Objective:Molecular cloning of human thrombopoietin(TPO)cDNA,Methods:RT-PCR,molecular cloning and DNA sequencing etc.methods were used.Results:Human thrombopoietin cDNA was cloned from fetus liver by RT-PCR techniques,The full lenghth cDNA including the sequence cod-ing signal peptide was obtained,and l062 b.p.of cDNA were sequenced by Sanger's method.Conclu-sions:A full lenghth of human TPO cDNA was obtained and subcloned.Expression of TPO and func- tional check is under way

目的:克隆人血小板生成素基因,方法:采用RT一PCR的方法。结果:从人胎肝中克隆出全长的血小板生成素(TPO)基因,并亚克隆至PUCll8载体中。该cDNA全长1062BP,编码353个氨基酸,经序列测定与Bartley等人报道的完全一致[1]。结论:已获得人血小板生成素基因,此项工作为基因工程生产TPO奠定了良好基础。

Objective: To get functional fragment of thrombopoietin gene. Methods: mRNA of hepatocyte was extracted as template, functional fragment of thrombopoietin gene was amplified by PCR, cloned into pUC19, and sequenced according to T7 DNA sequencing Kit. Results: The 480 bp functional fragment of thrombopoietin gene was obtained by PCR and determined by sequencing. Conclusion: The thrombopoietin gene has been cloned.

Monoclonal antibodies against murine CD4 or CD8 molecules were employed to directly stain the mononuclear cells from the immune tissues, they were analysed by flow cytometry. Results Mice treated with null plasmid or saline were made as controls....

Results Mice treated with null plasmid or saline were made as controls. It was discovered that with the delivery of TPO cDNA, both mature and the immature T lymphocytes in bone marrow increased in frequencies after a transient declination. Immature T lymphocytes increased in thymuses, while mainly mature T lymphocytes increased in spleens and blood circulation. Significantly among them was that CD4+ T subsets selectively proliferated in the process. They had formed the characters around 2 weeks after gene transference and remained such ratios for at least 2 weeks. Conclusion TPO be an immunoregulator and the cytokine over production in gene therpay mice is due to the activation of T lymphocytes, especially the helper subgroups.

目的 探讨TPO(血小板生成素)基因导入对T 细胞增殖和分化影响的规律。 方法 将编码人TPO 的质粒60μg 用脂质体包裹后,经肌肉途径转导入Balb/c 小鼠体内。用CD4和CD8的特异性抗体标记小鼠各种免疫组织中的T 细胞,借助流式细胞术分析它们的组成和变化。 结果 发现TPO 基因导入使(1)骨髓中的成熟和未成熟T 细胞数量在经过短期下降后全部大幅度增多;(2)胸腺未成熟T细胞增加;(3)脾脏和血液循环中成熟T 细胞增多,特别是在血液中CD4阳性细胞的增加最为明显。这些特点大约在基因导入后两周内形成,并持续至少两周以上。 结论 提示TPO 是一种免疫促进分子,TPO 基因导入后小鼠血液中多种淋巴因子的升高与它过量表达对T 细胞的刺激有关。
