Phenomena of autophage in pancreatic acinar cells of acute heamorrhagic - necrotizing pancreatitis (AHNP) rat was studied. Ultrastructure. CMP ase cell chemical and morphometric analysis of acimar cell in AHNP were investigated be fore and after treatment with target liposome of emodin. Results showed autophagic structure increased sig-nificantly and the ratio of primary lysosome was higher at 3hr after induction of AHNP as compared with control group.At 6 hour, the number and size of autophagic vacuoles were increased. Majority of the lysosome belonged to the sec-ondary type as evidenced by positive CMP ase reaction. Inclusion bodies in the vacuoles consisted of either organelles ormyeloid structure. In contrast, autophagic structures decreased in number and size and many belonged to the primarytype in target liposome of emodin group at 6 hr. Conclusion: Emodin can prevent redistribution of lysosomal enzymesand thus decreases the chance of zymogen activation. This suggests the stablizing effect of Emodin on lysosomal mem-brane.

本研究观察导向大黄素脂质体对急性重症胰腺炎大鼠胰腺腺泡细胞中自体吞噬现象的影响。用超微结构、CMP酶细胞化学和形态计量技术观察重症胰腺炎大鼠胰腺腺泡细胞的自体吞噬现象,并观察应用导向大黄素脂质体后的自体吞噬现象。结果示发病3小时后,细胞装内自噬体和自体吞噬泡增多,初级港酶体比例高于次级溶酶体。6小时后,自体吞噬泡数量及体积明显增多增大,内含物为细胞器或髓鞘样结构,CMP酶阳性反应部位以次级溶酶体为主。导向大黄素脂质体治疗6小时后,自体吞噬结构明显减少,CMP酶阳性反应区域多为体积较小的初级溶酶体。表明导向大黄素脂质体可能通过稳定溶酶体膜的作用来阻止溶酶体酶的重新分布,从而减少溶酶体酶和消化酶原在分泌自噬泡中接触激活的机会,也大大群低了自体吞噬泡的出现率。
