The ~(14)C-labelled and ~(125)I-labelled 4-iodo-phenoxyacetic acid (4-IPOA) were

Qualitative and quantitative examination of nucleic acid content in phloem exudate from pumpkin (Cucurbita moschata) and towel gourd (Luffa cylindrica) was conducted by chemical analysis in combination with electron microscopy. It was shown that when nucleic acid preparations were examined by two-wavelength and double beam spectrophotometer, the high and low absorption peaks were at 258 nm and 230 nm, respectively, (Fig. 1) which was in accord with typical ultraviolet absorption spectrum of nucleic acids. Qualitative and quantitative examination of nucleic acid content in phloem exudate from pumpkin (Cucurbita moschata) and towel gourd (Luffa cylindrica) was conducted by chemical analysis in combination with electron microscopy. It was shown that when nucleic acid preparations were examined by two-wavelength and double beam spectrophotometer, the high and low absorption peaks were at 258 nm and 230 nm, respectively, (Fig. 1) which was in accord with typical ultraviolet absorption spectrum of nucleic acids. DNA and RNA contents in exudate from pumpkin were 60-238 μg/ml and 30-80 μg/ml, respectively, while the contents in exudate from towel gourd were 30-120 μg/ml and 20-40 μg/ml, respectively. T, G. C, and A bases of DNA preparation from pumpkin were, after hydrolysis, separated satisfactorily by HPLC (Fig. 9). A clear electron micrograph of DNA molecule was obtained (Fig. 4) and the presence of nucleic acid was also shown through cytochemical examination (Fig. 3). The results described above showed that nucleic acids are present in phloem exudate from pumpkin and towel gourd. They are probably involved in phloem transport and may migrate along with the exudate flux.
