The materials were first fixed in different fixatives, embedded in paraffin, sectioned serially into 4fi thickness and stained with dyes of safranin and orange G, Heidenhain's iron haematoxylin combined with Mallory's connective tissue stain, buffered Magn-Lillie eosin methyl blue, Feulgen's reaction and methyl green-pyronin. Nucleolar extrusion was observed in all of these materials. Cells of nine rabbits stained with Heidenhain's iron haematoxylin were counted according to the cells with and without extruded nuckoli. The cells with extruded nucleoli constitute about "21.8% of the total number of paranchymal cells and most of them belong to chromophobes and least to acido-philes. The extruded nucleoli were situated in various parts inside or outside the cells or even within sinusoids. The entrance of the extruded nucleoli into sinusoids suggests that the nucleolar extrusion may have some relation to the secreting activity of these cells.

As indices for replication, we used the percentage of occurrence of polyhedrosis in ceils, TCID 50 assay, infection test in vivo and ultrastructural changes in infected cells. We found that AcNPV still maintained normal morphology and infectivity in cell line IPLB-SF-21 AE after 50 passages. It is thus possible to replicate Baculovirus in vitro for a long time without alteration of its characteristics. However, its morphology changed and its infectivity declined steadily following sequential passages in 21 AEC, 806 and 805 cells, but were rapidly restored after 5 passages in 21 AE cells.

In this study, the colony forming-units (CFU) were performed to obtain the adhesive index (AI) of Campylobacter jejuni (C. jejuni) strain CJ-S131 under different conditions. Factors affecting the adhesion were examined to probe the suitable conditions of the interaction between the CJ-131 Strain and the human epithelial cell monolayer in vitro. The results showed that the adherence was owed to the concentration of bacterial suspension, the type of cells, the temperature, pH, the time of incubation of bacteria with cells and bacterial motility. The multiple variable regression analysis by the computer revealed that both the concentration of bacterial suspension and the epithelial cell lines were significant factors related to the values of AI. Maximal adherence occurred in Hep-2 cell selected, approximately 1×10~7 CFU/ml bacterial suspension, at 37℃ and pHT.2, 4h after the bacterial suspension had been added to the monolayer of Hep-2 cell.

采用CFU法研究空弯菌在体外对单层上皮细胞的粘附,探讨空弯菌体外粘附的影响因素及最适条件。结果表明:空弯菌浓度、细胞种类、二者共同孵育温度,作用时间、细菌悬液的pH值及细菌动力等因素,都能影响空弯菌的粘附指数(AI)。多因素回归分析显示,细菌浓度、细胞种类与AI呈高度相关。正交试验提示:以Hep-2细胞为模型,细菌浓度约1×10~7CFU/ml,细菌与细胞在37℃、pH7.2的条件下,作用4小时是CJ—S131菌株体外粘附的最适条件。
