在普通CTAB法与SDS法提取RNA的基础上,从去除多糖和DNA污染两个方面进行优化,筛选出适于富含多糖类葡萄风信子花瓣RNA提取的方法,经检测OD260/280值在1.8~2.0之间,电泳28S条带和18SrRNA条带都很清亮,比值约1.5。对改进的CTAB法提取的总RNA进行RT-PCR能扩增出所需目的条带。有效去除多糖类物质污染对于富含多糖类花瓣组织RNA提取很重要。
