The experiment results of the rapid detection Escherichia coli in pharmaceutical products by the MUG-Indole method is reported in this paper. This is a new method based on technology using a MUG and Indole in single tube for the identifications and detection of Escherichia coli. The method effectively detected Escherichia coli within 24h.In case of positive results shown with MUG and negative with Indole or negative with MUG and positive with Indole. The proposed MUG-Indole method required further addtional IMViC tests should be carrid out. The test scheme has been proved to be successful in reducing false-positive and false-negative results to a minimal acceptable limit levels of the detection of Escherichia coli (<1%false-negative rate) .

本文报道用MUG-Indole检定大肠杆菌方法,在24h内能将94%~97%的大肠杆菌检出。对MUG阳性、Indole阴性或MUG阴性、Indole阳性的菌株,辅以EMB琼脂或麦康凯琼脂平板分离,挑选可疑菌落,做IMViC试验,使假阳性和假阴性降到最低水平。本法比传统的生化试验检定大肠杆菌法快速、简便、准确、无主观性。

In this paper,we report a comparative study on the survival and mutation of Escherichia coli in 35 pharmaceuticals by MUG-Indole(M-I)method and the method issued by the Health Ministry of China.The results showed that detection rate sensitivity,and accuracy with the above methods were similer on the whole.The contaminated E.coli in pharmaceuticals can be detected within 24 hours after enrichment culture by M-I method.M-I method is more significant in its simplisity and accuracy in comparison with the ministry-issued...

Methylumbellyferyl β D glucuronide(MUG)is firstly used to detect E.coli in pharmaceutical products.More than 6 % of the E.coli,however,do not be checked out with MUG.The MUG-Indole(M—I)assisted with IMViC test,a new method,could once check out the E.coli with positive MUG and positive Indole.Also,the new method could check out the E.coli with negative MUG and positive indole.So,the miss check problem of false negative and false positive results could be successfully solved.This method has the characteristics of quickness,simplicity and accuracy. The comparative experiments of both this new method and Chinese Pharmacopia to detect E.coli,have been done collaboratively by 20 provincial and municiple institutes for drug control and some pharmacy manufactories.The results indicated that:1.In 1576 batches of Chinese traditional patent medicines,45 batches were check out E.coli and 1494 batches were not checked out E.coli by both methods.The consistence of both methods to check E.coli was 97 7 % 2.It has been extensively analyzed for the unconsistence results of both methods,e.g.MI test ++ IMViC test ++-+ and MI test -- IMViC test -+--.3.When the results of IMViC test were ++-- or -+--,it might be other bacteria of non-E.coli in Escherichia.

首次应用4甲基伞形酮βD葡萄糖醛酸苷(4MethylumbeliferylβDglucuronide,简称MUG)检定药品中大肠杆菌获得成功,但有6%以上的大肠杆菌MUG为阴性反应,故提出MUG-Indole法,可将MUG阳性、Indole阳性的大肠杆菌一次检出;对MUG阴性、Indole阳性或MUG阳性、Indole阴性的大肠杆菌辅以EMB及IMViC试验,成功地解决了假阴性和假阳性的问题,提高了本法的准确性。
