A segment of autologous sciatic nerve was transplanted into the 17/18a border

将自体坐骨神经移植到成体金黄地鼠视皮层17/18a 交界附近,或靠近枕部大脑的中缝处。手术42~131天后,在移植的外周神经断端,给以 HRP(浓度50%,溶于生理盐水中),观察HRP 逆行标记的再生视皮层神经元。发现外周神经移植法可诱导成体金黄地鼠视皮层神经元再生;各种大小不等、形状各异的神经元,都可再生出纤维到移植的外周神经中,长达10mm;再生神经元分布于靠近移植神经的皮层Ⅱ~Ⅵ层中;某些再生的神经元胞体,较正常细胞明显增大,这与金黄地鼠视网膜神经节细胞轴突再生的情形类似,但金黄地鼠视皮层神经元再生能力远不如视网膜神经节细胞。本实验用荧光双标记法,证实皮层视区胼胝体神经元是可以再生的。用“预先溃变”和“额外损伤”法,未能导致视皮层再生神经元数目的增加。本实验结果还表明,外周神经移植法可诱导金黄地鼠视皮层神经元再生,局部环境因素是影响哺乳动物中枢神经元再生能力的重要原因之一。

All the rats. under either anaesthesia. were injured by partial decortication bilaterally in the parietal region. The wound,3×3mm in surface area and 1mm in depth, was created with a small spatula. A piece of gelfoam (2×2×1mm3)was soaked with 25μl of BWTE. It was placed in the wound cavity 4 time over a peried of two months.In the control rats Hank's solution was used in stead of BWTE in equal quantity at the same time. 60 days after the operation, the brains were removed out and a 2mm thick block was taken along the coronal plane to inciude most of wound area.The brain tissue was researched quantitatively and qualitatively under light and electron microscope and immunohistochmisiry.

本研究的目的是探讨神经元的再生。SD成年雄性大白鼠38只,随机配对为实验组和对照组。大鼠经切除双侧大脑皮质顶叶下正中部分(4mm×3mm×1mm),用大约2mm×2mm×1mm海绵胶(gelfoam)粒浸入脑损伤组织提取液(BWTE)中,术时将上述海绵胶粒放入大脑皮质损伤腔内,2个月内加入BWTE4次。对照组用上述方法加入等量的Hnnk’s液,术后60d取材。取含损伤腔的大脑皮质,用Nissl染色,辅以神经丝抗体免疫染色以及电镜结合体现学定量方法分析,结果实验组与对照组相比发现BWTE组可明显促进损伤皮质神经无的存活(P<0.001)。实验组出现较多胞体增大的神经元(P<0.02),且多具有粗大的顶树突。体视学定量方法分析结果是:神经元的线粒体体密度、数密度、面密度以及粗面内质网的面密度均较对照组增加(P<0.05)。提示BWTE对成年大鼠损伤的皮质神经元具有神经营养活性,可能含有神经营养因子(neuronotrophicfactors,NTFs)。

将从3周龄大白鼠腓骨肌中提取的22、35kD的肌肉源神经诱向因子(MNTF),用1×1mmPhast凝胶块运载后放入游离周围神经移植与脊髓连接处。结果表明:术后4周,实验组游离神经与脊髓连接处无明显空洞反应,大量神经纤维长入神经内,再生的有髓纤维为对照组的10.5倍(P<0.01)。HRP示踪发现实验组有1480.3±423.5个阳性神经元,而对照组只有20.3±6.5个神经元(P<0.01)。显然,MNTF具有增强脊髓及神经根背节神经元,尤其是脊髓运动神经元再生的能力,并可能有营养移植神经中雪旺氏细胞的作用。这对脊髓损伤修复具有一定的意义。
