AIM:To find a suitable microenviroment for improved induction of plasma cells from murine embryonic stem(ES)cells,we explored the roles of cytokines and primary bone marrow stromal cells in the development of ES-D3 cells.The process was divided into three stages.First stage(0-4 day):adding SCF(stem cell-factor) ,TPO(thrombopoietin),IL-6,IL-3,erythropoietin,G-CSF into methycellulose culture,ES-D3 cells died four days later,ES-D3 cells developed into embryoid bodies and they grew well in the presence of 2-Mercaptoethanol...

AIM:To find a suitable microenviroment for improved induction of plasma cells from murine embryonic stem(ES)cells,we explored the roles of cytokines and primary bone marrow stromal cells in the development of ES-D3 cells.The process was divided into three stages.First stage(0-4 day):adding SCF(stem cell-factor) ,TPO(thrombopoietin),IL-6,IL-3,erythropoietin,G-CSF into methycellulose culture,ES-D3 cells died four days later,ES-D3 cells developed into embryoid bodies and they grew well in the presence of 2-Mercaptoethanol (2-ME) only.Second stage(4-7 dyas):the embryoid bodies were trypsined,then transferred into secondary methylcellulose culture,cells grew slowly when adding SCF and TPO, but they(86%)developed quickly into the colonies of CD34 + cells in the presence of conditional medium(first stage of liquid culture,adding 2ME only).Third stage:CD34 + cells developed into CD34 - CD38 + cells when transferred onto prepared bone marrow stromal cells in the presence of IL-6 and SCF,they looked like plasma cells by cell morphology,and immunoglobulin test was positive .CONCLUSIONS:Cultured in a suitable microenviroment,ES-D3 cells can developed into plasma cells.cytokines were not needed in the early development of ES-D3 cells,conditional medium of the first stage of liquid culture could accelerate them to develop into hematopoietic cells,and the primary bone marrow stromal cells play a main role in the induction of plasma cells.

目的:建立胚胎干细胞体外定向诱导分化为浆细胞的体系,为研究造血发生的基因调控及B淋巴细胞发育过程奠定基础。方法:分阶段培养,第一、二阶段使用半固体培养,分组加入细胞因子、2-巯基乙醇和胚胎细胞条件培养液,用流式细胞仪、免疫组化及Wright’s染色鉴定细胞,第三阶段移入骨髓基质细胞滋养层。结果:第一阶段:加入SCF、TPO、IL-6、IL-3、Epo、和G-CSF,4d后细胞大片坏死,仅加入2-巯基乙醇可形成胚胎体。第二阶段:加入SCF和TPO,细胞增殖缓慢,再加入条件培养液,发育3~4d可形成CD34+的细胞集落。第三阶段:移至骨髓基质细胞饲养层后,发育为浆细胞。结论:胚胎干细胞在体外可定向分化为浆细胞。胚胎干细胞在早期发育不需要细胞因子,胚胎细胞的条件培养液可促使其向造血细胞发育,骨髓基质细胞饲养层可使其向浆细胞分化

AIM: To study cellular and molecular mechanisms of cardiac development associated genes ex- pression and its function during early stage cardiomyogenesis. METHODS: (1 ) Mouse embryonic stem cells (ESC) line D3 culture. (2) Inductive culals of ESC differentiated into cardiac myocytes in vitro.(3) Identification of ESC -derived cardiac myocytes: RNA isolation; synthesis of specific primer and RT - PCR; Label of RT - PCR products with [α - 32P] dATP as probes, purifyed by sephadex G - 50 columns, determined the yield of DNA. RNA dot hy- bridization. RESULTS: 80% of ESC differentiated into cardiomyocytes by improved conditional medium. Cardiomy- ocytes contraCted in a synchronous manner. The results of RT - PCR and RNA blot showed that cardiac genes were expressed abundantly and specifically during the early cardiomyogenesis. CONCLUSIONS: ESC were able to be dif- ferentiate into cardiomyocytes. Different concentrations and components of RA, DMSO and FCS affected ESC car- diomyogenesis in de. The optimal result obtained was from the conditional medium, a mixturce of 2 nmol/L retinoic acid (RA), 0.6% dimethyl sulfoxide (DMSO) and 20% fend calf serum (FCS).

目的:用胚胎干细胞(ESC)体外诱导分化为心肌细胞,从分子水平上研究早期心脏发育相关基 因及其功能。方法:(1)胚胎干细胞的培养。(2)胚胎于细胞的分化培养。(3)被分化的心肌细胞鉴定:RNA的 提取;心脏特异性引物的合成和RT-PCR反应;探针标记、纯化和比放射活性测定;RNA斑点杂交。结果:用最 适条件培养液对FSC定向诱导分化,可使80%以上的ESC分化为心肌细胞。心肌细胞以同步的方式进行收缩。 反转录PCR和斑点杂交的结果显示:心肌在早期发育就开始表达其特异性基因。结论:胚胎干细胞体外能诱导 分化为心肌细胞。胎牛血清、二甲基亚砜和维甲酸的浓度及它们之间的组成不同,对ES细胞定向诱导为心肌 细胞均有影响。最佳诱导条件是用2nmol/L维甲酸、0.6%二甲基亚砜和20%胎牛血清组成的条件培养液。

Objective:To study the capacity of differentiate committedly of recombinant human thrombopoietin(rhTPO) on megakaryocyte progenitor cells in peripheral stem cells(PBMNC). Objective:To study the capacity of differentiate committedly of recombinant human thrombopoietin(rhTPO) on megakaryocyte progenitor cells in peripheral stem cells(PBMNC). Methods: PBMNC mobolized by chemotherapy and GCSF were cultured in suspension and colony culture, study the effects of rhTPO along and/with IL3,IL6,SCF.Results: After culture, the MNC counts of PBMNC were expansed up to 1.73±0.49 folds by TPO along, a combinantion of IL3,IL6,SCF expands the MNC 4.20±1.14 folds, and a combinantion of IL3,IL6,SCF and TPO expands the MNC up to 5.43±1.27 folds. TPO along or a combinantion of TPO with other cytokines result a high percentage of CD41a+ cells,with TPO, CD41a+cells were 11.07%±5.23% before culture and 19.17%±6.26% after culture. After culture, the CD41a+cells were expansed up to 3.52±1.18 folds by TPO along, and up to 5.32±1.79 folds by IL3,IL6 and SCF, 6.94± 2.19 folds by IL3,IL6,SCF and TPO. Conclusion:TPO can inducing megakaryocyte progenitor cells differentiate committedly, and IL3,IL6 and TPO can synergized with TPO.
