Role of B cells in growth of mul-tipotential progenitor cells was extensi-vely studied when we had reported theaugmentative effects of B cells on pro-liferation of CFU-S and CFU-GM,andthe stimulating activity in the condi-tioned medium of B cells.B cells weresuccessfully isolated from the mousespleen by means of free-flow electropho-resis(Elphor Vap-11)as usual.Theisolated B cells samples(IBC)containing93.5±1.1% of Anti-IgM.G.A-FITCpositive cells and 0% of Anti-Thy 1.2-FITC positive cells.Colonies derivedfrom CFU-mix increased to 20.7±2.8in the co-culture of BMC and IBC ascompared with the number of CFU-mixcolonies 4.4±0.3 and 2.1±0.2 respec-tively in the seperate cultures of BMCor IBC alone with the same amount asin co-cultivation.Number of p-BFU-Ecolonies in co-culture of IBC and BMCwas extremely higher than that in thecontrol. Colonies of CFU-mix markedly in-creased in the presence of conditionedmedium of a transformed B cell line(M12.4.1-CM)3.9 times as many asthat in the control whereas no colonyat all could form in the culture addedwith M12.4.1-CM only but withoutaddition of any conventional CSF-mix.This result was consistent with the sug-gestion that in the M12.4.1-CM therewas just activity of co-CSF-mix butnot CSF-mix. It was shown by cytospin for sing-le mix-colony analysis that numbers ofCFU-GEMm and p-BFU-E were enhan-ced significantly up to 25.0 and 55.0%respectively in the cultures with M12.4.1-CM whilst just 6.7 and 37.8% inthe control without M 12.4.1-CM. A notable result happened whenIBC were cocultured along with 1000-rad-irradiated BMC that CFU-mix emer-ged up to 19.3±1.0 per dish whileCFU-mix colonies could just be enu-merated as 2.1±0.2 and 0±0 per dishrespectively in culture of IBC or 1000-rad-irradiated BMC seperately in theconventional culture system. Our data showed the influence ofdifferent activities in culture on various ??types of colonies which formed in theculture of IBC alone,e.g.with ML-CM more CFU-GEMm and CFU-GMmemerged,however,more p-BFU-E form-ed when adding ML-CM along withLung-CM. Colonies derived from CFU-mixwas reducing when various dilutions ofanti-IgM serum and complement wereadded in the culture of IBC.A linearcorrelation was shown between the nu-mber of CFU-mix in culture countedand the amount of anti-IgM serum ad-ded. What remains obscure is the possi-bility of rejuvenation of a small part ofB cells to multipotential progenitor cellsunder certain microenvironment.It is tobe clarified if minority of B cells arethe candidates of hemopoietic progenitorcells or B cells and CFU-mix have thesame property of surface electron to mixup during free-flow electrophoresis.

小鼠骨髓细胞在体外培养中,加入用流式自由电泳法分离所得的高纯度正常B淋巴细胞,可使多向祖细胞(CFU-mix)集落增加至5倍;加入小鼠B淋巴瘤细胞株的条件培基(M_(12.4.1)-CM)时,CFU-mix数也可增加至4倍。单集落形态学分析结果表明M_(12.4.1)-CM可加强CFU-GEMm及p-BFU-E等早期造血祖细胞的增殖与分化。小鼠高纯度B细胞样品在体外培养中加入1000 rad照射的骨髓细胞可出现CFU-mix集落,如果再加入适量的小鼠肺条件培基,则CFU-mix数量比对照大15倍,其集落性质为CFU-GEMm,GMm及p-BFU-E。在此培养中加不同稀释度抗小鼠IgM血清,结果CFU-mix的产率与抗IgM血清的浓度成直线反比关系,当加入1:10抗小鼠IgM血清时,CFU-mix为0。作者假设在一定培养条件下,IgM阳性的部分B细胞可返祖转化为CFU-mix。
