It was found that the grafts of arenal medulla were able to provide a substantial restoration of the dopaminergic neurotransmitters in denervated striatum and dorrect the motor asymmetry of the rats with Parkinson's disease. Our findings suggest that intracerebral transplantation of adrenal medulla grafts in an efficient therapy for Parkinson's disease and renervation between the recipient and the grafts in the possible basic mechanism of the recovery.

采用高压液相色谱──电化学检测法测定纹状体内单胺类递质多巴胺及其代谢产物3,4-二羟基苯乙酸、去甲肾上腺素,以研究肾上腺髓质脑内移植后帕金森病(PD)鼠纹状体内部分神经生化改变。结果发现肾上腺髓质脑内移植可以基本矫正PD鼠异常旋转行为、弥补单胺类神经递质缺乏。研究结果提示肾上腺髓质脑内移植后可能与宿主神经组织形成神经再支配,肾上腺髓质脑内移植是治疗PD的较有效的手段。

脊柱侧弯被定义为脊柱相对正常中心线明显地弯曲伴椎体异常旋转。它在冠状面上出现不对称弯曲,在横断面上有异常旋转。所以一个合格的脊柱侧弯动物模型应具有人脊柱侧弯的特征性变化,并且简单易行,有可重复性。我们是通过单纯于兔肩胛骨与同侧股骨大转子间连以钢丝,使脊柱发育成侧弯且进行性加重椎体旋转,出现三维结构变化但不直接损伤脊柱,3组幼兔固定后3、4、5个月分别用过量异戊巴比妥钠予以处死,观察脊柱结构变化:顶椎出现楔形变并旋转;骨盆倾斜;椎间盘髓核移向凸侧;凹侧椎体生长骺板变薄,柱状增殖细胞层变得紊乱;凸侧椎板变宽而短,对侧窄而长。横断面上顶椎前后径在凹侧增宽,骨小梁密集。实验结果表明凹侧椎体生长骺板受到超高的压力而凸侧骺板受力相对小。因此凹侧骺板由于逐渐增加的超高力的作用从而使纵行生长受到抑制,椎体楔形变,脊柱侧弯进行性加重。

Objective: To observe whether engineered MN9D/GDNF cells stably expressing

目的:观察稳定表达GDNF的MN9D/GDNF工程细胞对6羟基多巴胺损毁所致帕金森病(Parkinsondisease,PD)模型大鼠旋转行为有无治疗作用。方法:用6羟基多巴胺损毁大鼠单侧中脑黑质多巴胺能神经元、2周后腹腔注射阿朴吗啡诱导大鼠出现偏侧旋转行为的方法制备PD模型大鼠。通过脑立体定位技术,将稳定表达GDNF的MN9D/GDNF工程细胞注入模型鼠损毁侧纹状体,1周后观察阿朴吗啡诱导的旋转行为有无改善。结果:稳定表达GDNF的MN9D/GDNF工程细胞可明显降低PD模型大鼠的异常旋转行为,治疗作用达10周之久。结论:运用exvivo方法和防治兼顾的策略对于PD基因治疗的基础研究及临床应用具有重要意义。
