Objective To experiment the possibility of allodecellular extracellular matrix for the replacement of ureter. Methods Extracellular matrix was obtained from the ureters of 20 rabbits by decellularization process.60 experimental rabbits were used and a 4cm segment of left ureter was resected in each.In the experimental group of 20 rabbits,the resected ureter was replaced by the allo ureter ECM.In the first control group of 20 rabbits the resected ureter was replaced by allograft transplantation of untreated ureter.In the second control group of rabbits the resected ureter site was left untreated for autoregeneration.The neoureter segment was studied with electron microscopy and immunohistochemical procedures. Results In the experimental animals,transitional epithelial cells and monocytes could be seen in the ECM area and fully filled up the area after 3 weeks.No stenosis and hydronephrosis were observed on IVU 24 weeks after replacement of the resected ureter.The histological structure was not much different from a normal ureter.In the first control group with allograft transplantation,rejection and necrosis occurred 7~10 days after the transplantation.In the second conrol group of autoregeneration the studied area was embeded with fibrosis tissue.On the basis of the experiment,ureter ECM has been successfully used in clinical practice. Conclusions Alloureter ECM might be an ideal replacement material for ureter.

目的 寻求理想的输尿管替代材料。 方法 将20 只兔双侧输尿管脱细胞处理后制成输尿管细胞外基质(ECM) ;另60 只兔分为3 组:输尿管ECM 移植组( 实验组) ,对照组1 及对照组2 。 结果 实验组术后组织学检查显示1 周时ECM 边缘部腔内面已见移行上皮细胞,管壁有肌细胞长入。6 周时取材ECM 区已修复近正常输尿管组织结构。12 、24 周时IVU 检查;输尿管显影良好,通畅。对照组1 术后7 ~10 天,移植的异体输尿管均坏死。对照组2 输尿管缺损区6 周时均被纤维包裹而明显增粗。在实验取得满意结果的基础上成功应用于临床。 结论 异体脱细胞输尿管细胞外基质为理想的输尿管替代材料。

The cell scaffold complex was transplanted in nude mice. Scanning electron microscopy, HE staining and immunocytochemical staining of cytokeratin were used to evaluate the growth of cells on the collagen membrane. Results Transitional cells adhered to collagen membranes and grew at 2 h after implantation. On day 20 after transplantation in vivo , the transitional cells lined in continuous layers of 1 to 5 cell thickness at some parts of the complex. Conclusion Transitional cells isolated in this way are suitable seeding cells to construct urethra by tissue engineering. The cell scaffold complex can form an analog of normal urethra after transplantation in vivo .

目的 探索以膀胱移行上皮细胞为种子细胞构建组织工程尿道的可行性。方法 取兔膀胱移行上皮细胞 ,体外培养传代后作为种子细胞与自制胶原膜体外联合培养并行裸鼠体内移植 ,通过对复合物扫描电镜、HE染色及角蛋白免疫组化检测 ,了解细胞在材料上的生长情况。结果  2h后移行上皮细胞在胶原膜上贴附生长 ,复合物移植体内 2 0d后 ,移行上皮细胞分化成层 ,层厚 1~ 5层。结论 该方法获取的膀胱移行上皮细胞是构建组织工程尿道的良好种子细胞 ,与胶原膜构建的复合物在体内形成了尿道类似物 ,有望成为替代尿道的组织工程材料

To establish a protocol for isolating,culturing,proliferating urothelial cells,and to obtain large number of autologous urothelial cells for tissue_engineered substitutes.Urothelial cells were isolated from sample tissues by scraping or tissue explanting and were cultured in Karetinocyte_SFM.Morphology and expansion of the cells were observed.Cell_type specific proteins were analyzed by immunohistochemical methods.The results showed that urothelial cells presented a typical morphological feature of transitional epithelium.Cells could be passaged 10 times without a noticeable decrease in rate of cell proliferation.Immunohistochemistry showed that CKAE1/AE3 was positive and CK34βE12 was negative.Therefore,in this protocol,the isolation of urothelial cells are simple,the cells proliferate rapidly and can be passaged easily.The biological properties of urothelial cells have been kept and no signs of senescence were seen even after the 10th passage.

为建立尿路上皮细胞分离、培养、扩增的常规 ,获得构建组织工程学泌尿系器官所需的大量尿路移行上皮细胞 ,分别采用刮削法和组织块法分离出尿路上皮细胞 ,在角化细胞培养基 (Keratinocyte_SFM)中培养 ,观察细胞形态、扩增情况 ,并用免疫组化方法检测细胞类型特异性蛋白质。结果 :尿路移行上皮细胞生长良好 ,呈现出典型的移行上皮形态特征。细胞经10次传代后仍扩增迅速 ,免疫组化染色显示CKAE1/AE3 阳性、CK34βE12 阴性。提示该常规中细胞分离方法可靠 ,传代顺利 ,扩增迅速 ;尿路上皮细胞经10次传代仍保持其固有生物学特性 ,未见老化
