In certain part of China, especially in Guizhou and some provinces adjacent to Guizhou, uri -nary bladder tumor is a common disease in cattle and associated with a syndrome known as chronic enzootic hematuria.According to epidemialogical data the occurrence of the disease was associated with the geographic distribution of two species of bracken ferns,pteridium aquilinum and pteridium revolutum, especlally the latter.It seems that pteridium revolutum acts a more important part for the occurrence of bovine enzootic hematuria in China, although pteridium aquilinum has been considered as a causative factor of the disease by "many previous works in the world.Carcinogencity of pteridium revolutum was found first in the world by author and coworkers, and reconfirmated by other wrokers in China. A small amount of the plant (3-5g/day/kg of body weight) fed to cattle for a lon,5 time (2.5-3 years) produced urinary bladder tumors indistinguished from the naturally occurring bovine enzootic hematuria that developed in the bracken-fern-infested areas. Some other species, except goat and rabbit, are also susceptible to the carcinogenic action of pteridium revolutum. Orally administrationl of the plant induced urinary bladder tumors in guinea-pigs and both intestinal and urinary bladder tumors in various strain of rats, just like these animals reacted to pteridium aquilinum.

在我国的部分地区,特别是在贵州及与之相邻的一些省区,牛膀胱肿瘤极为普遍,且与一种被称为慢性地方性血尿症的综合症相关联。根据流行病学资料,该病的发生与两种蕨属植物——蕨(Pteridium aquilinum)及毛叶蕨(Pteridiumrevolutum),特别是毛叶蕨的地理分布有关。国内外已有不少人证实蕨为该病的病因,而我国牛地方性血尿症的发生似与毛叶蕨关系更为密切。最近笔者等首次在世界上证实了毛叶蕨的致癌性,并被国内其他工作者随后的工作重复证实。长期(2.5~3年)少量(3~5克/每日公斤体重)给饲毛叶蕨可诱发出与毛叶蕨密布区域自发性的牛地方性血尿症相同的牛膀胱肿瘤。除山羊及家兔外,其他种类的一些动物对毛叶蕨的致癌作用也是敏感的。经口投给毛叶蕨诱发豚鼠的膀胱肿瘤和在不同品系的大鼠中诱发出肠道与膀胱的肿瘤。

The changes of adhesiveness of BCG to a transitional carcinoma cell line 253J with low FN expression were investigated before and after FN cDNA transfection by lipofectinmin. The result showed that FN level was increased in the cells with more BCG adhered to. It suggested that FN expression plays a very important role in mediating BCG adherence to tumor cells.

为探讨纤维粘连蛋白(Fibronectin,FN)介导BCG抗膀胱肿瘤的机制,利用转基因技术将FNcDNA导入低表达FN的膀胱癌细胞系253J,观察转染前后[3H]Thymidine标记的BCG与肿瘤细胞粘附的变化。结果显示:转染后的肿瘤细胞FN表达水平明显增高;同转染前的肿瘤细胞相比可结合更多的BCG(P<0.001)。提示FN的表达在BCG与膀胱肿瘤的粘附中起着重要的作用。

Objective To clarify the influence of E cadherin on the invasive ability of bladder tumor cells. Methods 40 fresh bladder cancer tissues were investigated by matrigel invasion assay and direct sequencing of amplified E cadherin complementary DNA fragments. Results In 32 grade Ⅰ and Ⅱcases,E cadherin mutation was identified in only 2 while it was detected in 4 of 8 grade Ⅲ and Ⅳ cases revealing a positive correlation between E cadherin mutation and the differentiation of tumor cells.E cadherin alteration...

Conclusions E cadherin mutation can significantly influence the invasive ability of tumor cells.On the other hand,some tumors without E cadherin alteration may also have invasive ability denoting that other mechanisms should be consi dered .

目的观察Ecadherin基因突变对膀胱癌侵袭力的影响。方法对40例新鲜膀胱癌标本分别完成体外matrigel穿膜侵袭实验和Ecadherin基因的序列测定。结果病理分级Ⅰ、Ⅱ级者32例中仅有2例出现Ecadherin基因突变,而Ⅲ、Ⅳ级者8例中有4例出现突变,说明Ecadherin变化与肿瘤细胞分化之间有明显的相关性。本组6例标本出现Ecadherin结构改变,其中4例穿膜侵袭实验阳性,而34例未发现Ecadherin基因改变的标本中,只有7例穿膜侵袭实验阳性。结论Ecadherin基因改变对肿瘤细胞的侵袭能力有明显影响。未发现Ecadherin基因突变的标本也可能具有侵袭能力,说明在膀胱肿瘤的侵袭过程中存在其它机制。
