To investigate the effect of HPS on bladder cancer line EJ cell proliferation was tested by 3-(4,5-dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide (MTT) assay.The results showed that HPS hasn't enhanced effect for LAK cell proliferation.When simultaneously applied with HPS cytotoxicity of LAK cell or PBMC were enhanced agaist EJ cell and BTC from the patients.

用MTT法观察了红芪多糖(HPS)对LAK细胞的影响,结果表明HPS无明显的促进LAK细胞扩增的作用,但与LAK细胞或PBMC合用可显著增强对膀胱肿瘤细胞株EJ和原代肿瘤细胞的杀伤作用。

OBJECTIVE:To study the in vitro and in vivo killing effect of Newcastle Disease Virus(NDV) on bladder carcinoma cells.METHODS:Bladder carcinoma mice model(BST739) was adopted and cells culture,cells counting,light microscope and electron microscope technique were used.RESULTS:Result of in vitro trial showed that significant killing effect on bladder carcinoma were observed in all 3 groups of NDV,BCG,and MMC and virus particles were found in cytoplasm by electron microscope.Results of in vivo study showed that some tumor cell killing effect were observed in all 3 agents of NDV,BCG,and MMC and some inhibitory effect on bladder tumor were observed in NDV group when administered by intra tumor and opposite tumor injection but obvious lymph cell invasion was observed in NDV group in comparison with control group.CONCLUSION:NDV has some in vitro and in vivo killing effects on bladder tumor cells.

目的:研究鸡新城疫病毒(NDV)对膀胱癌细胞在体内、体外的杀伤作用。方法:采用了细胞培养和计数、光镜、电镜观察的方法及小鼠膀胱肿瘤模型(BST739)进行研究。结果:体外实验中,NDV组、卡介苗组及丝裂霉素C组对膀胱癌细胞均有明显的杀伤作用;用透射电镜观察NDV作用后的膀胱癌细胞,发现胞浆中有病毒颗粒。体内实验证明,NDV、丝裂霉素C和卡介苗对膀胱癌细胞都有一定的杀伤作用;在瘤内和瘤对侧注射NDV对膀胱肿瘤生长都有一定的抑制作用,且瘤内注射的效果远好于瘤对侧注射组。瘤内注射NDV组与对照组相比有明显的淋巴细胞浸润。结论:NDV对膀胱癌细胞在体内和体外均有一定的杀伤作用。

Purpose:To study the effect of mitomycin-c retention in urinary bladder by intravesical instillation of HPC-MMC. Method:20 patients with urinary bladder tumor were observed in the present study, ten of which were administered with 1 % HPC-MMC instilled into the urinary bladder and the other ten were administered conventional aqueous mitomycin-C after TURBT or partial cystectomy. Purpose:To study the effect of mitomycin-c retention in urinary bladder by intravesical instillation of HPC-MMC. Method:20 patients with urinary bladder tumor were observed in the present study, ten of which were administered with 1 % HPC-MMC instilled into the urinary bladder and the other ten were administered conventional aqueous mitomycin-C after TURBT or partial cystectomy. MMC levels in urine were determined by high-pressure liquid chromatography and the pharmacokinetics were compared in both group. Result: After instillation, the HPC-MMC remained significantly longer within the urinary bladder than S-MMC. Side effects were not found in all cases. HPC-compounded anticancer drugs might be therapeutically more useful for bladder tumor. Conclusion .-Combined intravesical instillation of the MMC and HPC could keep longer MMC in urine and should be recommended for clinical use.

目的:探讨粘膜粘附缓释剂羟丙基纤维素(HPC)与丝裂霉素C(MMC)联合膀胱灌注对MMC在膀胱中保留的效果。方法:20例膀胱肿瘤患者分为HPC—MMC联合组和单纯用MMC(S-MMC)组,行TURBT或膀胱部分切除术后分别用1%HPC—MMC和S-MMC行膀胱灌注,采用高压液相法检测尿中MMC浓度,并对比研究两者尿药动力学改变。结果:HPC—MMC组膀胱灌注后,MMC在膀胱尿中的滞留时间、滞留量明显优于S-MMC灌注组,临床应用未见副作用发生。结论:HPC与MMC联合灌注膀胱可使MMC持续保留,值得在临床推广。
