Aqueous humor samples were aspirated on the alkali burn 1,3,7,14 and 30 days,and the total number of white blood cells in the aqueous humor and their subset distribution as well as tumor necrosis factor-α (TNF-α) and NO~-_2/NO~-_3 were counted.
Decreased inflammation, lower protein concentration and fewer inflammatory cells in the aqueous humor and ED1+ cells in the iris-ciliary body were observed in the PRP-treated rats as compared with the untreated ones.
AIM: To study the inhibitory effect of pyrrolidine dithiocarbamate (PDTC) on the inflammatory reaction in an experimental proliferative vitreoretinopathy (PVR) model with laser flare cell meter (LFCM).
Results During the follow-up period, aqueous flare were observed in 30 eyes of group A (30.6%), compared with 42 eyes in group B(77.8%); aqueous cells were observed in 26 eyes of group A( 26.5%).
,43 eyes of group B(79.6%); iris posterior synechiae occurred in 17 eyes of group A(17.3%),compared with 24 eyes in group B(44.4%); posterior capsular opacity developed in 2 eyes of group A(2.04%),7 eyes of group B(12.7%).
On the operative day and on the 1st,3rd,7th and 14th days after operation,inflammatory reaction of anterior chamber in traumatic control group and operative group was measured and 0.2 mL aqueous humor was extracted from rabbits in three groups to count and classify cells; meanwhile,expressed level and dynamic changes of IL-1,IL-6 and TNF-α in cytokines of aqueous humor were measured with double-antibodies ELASA technique.
Results Different clinical signs was observed, 84.79±6.25pg/ml TNF-α was detected in the aqueous humor after the injection of 10 microgram lipopolysaccharide into the anterior chamber and 143.42±9.31pg/ml when 20 microgram lipopolysaccharide was injected, cell count,protein content and histologic grading varied with different sites injection and doses, and the difference between the test and the contol group was statistically significant.
TNF-induced ocular inflammation therefore differed from that reported for intravitreally injected endotoxin in terms of time course and the types of inflammatory cells in the aqueous humor.
