The interactions of OKY-1580 and OKY-046 with human serum albumin(HSA), the bindin affinity and the binding sites were studied. The ADP scavenger creatine phosphate/creatine phosphokinase (CP/CPK), or inactivation of cyclooxygenasc with aspirin failed to inhibit TMVA-induced activation. Thrombin-trcated, degranulated platelets was aggregated in response to TMVA in the presence of fibrinogen. Thus the platelet aggregation induced by TMVA was independent on the release reaction and the formation of TXA2. However, the TMVA-induced platelet activation was prevented by Mepacrine, Tetramethyl Pyrazine, a compund isolated from Chinese herbs, and SZ- 2 , a monoclonal antibody to human platelet membrane glycoprotein Ib. The actions of TMVA on human platelets were similar with that of PAF, suggesting that TMVA and PAF may share some common pathways of platelet activation.

During 5 days incubation, 5-HT was not diminished in the whole blood plus CSF. Levels of 6-keto-PGF_(1a), TXB_2, PGE_2 and LPO in spasmodic basilar and femoral arterial walls induced by applying 5-HT showed no biochemical change, but there are vacuolated deterioration on etectromicroscopic examination.
