AIM:To observe the effect of altitude hypoxia on γ-aminobutyric (GABA) content and prepro-somatostatin mRNA (PPS-mRNA) in the rat hypothalamus. METHODS: Using altitude hypoxia model,in situ hybridization and amino acid analyzer, the number of PPS-mRNA and GABA content in rat hypothalamus was determined. RESULTS:After altitude hypoxia, the contents of GABA in hypothalamus and the number of PPS-mRNA neurons in periventricular nucleus (PeVN), paraventricular nucleus (PaVN) and arcuate nucleus (ArcN) increased significiantly. Bicuculline, a GABA receptor antagonits, could enhance PPS-mRNA expression evoked by altitude hypoxia, but had no effect on GABA content. CONCLUSION: Altitude hypoxia can induce neurotransmitter imbalance of hypothalamus.

目的 :观察高原低氧大鼠下丘脑前生长抑素原 (PPS)mRNA和γ -氨基丁酸 (GABA)含量变化。方法 :应用大鼠高原低氧模型及原位杂交技术和氨基酸测定法 ,检测下丘脑前PPS -mRNA神经元数量和GA BA的含量。结果 :高原低氧组大鼠下丘脑GABA的含量明显增多 ,室周核、室旁核、弓状核PPS -mRNA阳性神经元数目增加 ;而GABA受体拮抗剂荷包牡丹碱使高原低氧大鼠下丘脑PPS -mRNA阳性神经元数目增多更为显著 ,但对GABA含量的增加影响不大。结论 :高原低氧可导致下丘脑神经递质代谢紊乱

Aim and Methods:Contents of glutamate (Glu),asparate (Asp) and expression of prepro somatostatin mRNA (PPS mRNA) in rat hypothalamus were measured by using imitated altitude hypoxia rat model, amino acid analyzer and in situ hybridization technique.Results: After rats were subjected to altitude hypoxia,contents of Glu and Asp in hypothalamus and PPS mRNA expression in periventricular nucleus (PeVN),paraventricular nucleus (PaVN) and arcuate nucleus (ArcN) were increased significiantly.Ketamine, a NMDA receptor antagonist,could decrease the number of PPS mRNA neurons in rat hypothalamus evoked by altitude hypoxia, but had no effect on Glu and Asp contents evoled by altitude hypoxia.Conclusion:It is suggested that somatostatin maybe paticipate in altitude hypoxia reaction,Glu can enhance PPS mRNA expression through NMDA receptor.

Bovine pericardium is mainly composed of collagen which has many good properties of extracellular matrix. The purpose is to find out one mehtod that presents not only the degradation regularity of material but also minimum antigenicity, so that bovine pericardium can be used as a well degradable guide tissue regeneration material. The results of the experiments showed that the bovine pericardium treated with ethanol is more appreciated than that treated with epoxy cross linking agent or glutaraldehyde.The mensuration of protein and special amino acid may be a useful base of constructing a mathematic model for further researches on the rate of degradation of bovine pericardium.

牛心包材料是一种以胶原蛋白为主要成分的天然生物材料。为了研究牛心包作为组织工程材料的可能 ,本实验通过采用失重法、蛋白质检测法及氨基酸测定法定量地研究和对比了几种不同改性方法处理过的牛心包的体外降解规律 ,力求找到一种改性方法使材料既有可控的降解性 ,又有较小的抗原性 ,以使牛心包成为一种良好的可降解性 GTR材料。研究结果显示 ,用乙醇封端处理的心包相对于戊二醛及环氧交联处理的心包 ,在降解性方面更易满足可降解 GTR材料的要求。蛋白质检测法及氨基酸测定法也为以后建立控制心包降解速率的数学模型奠定了良好基础
