AIM: To investigate the pathway net of visceral receptors and probe into the correlation of pathway of gastric adventitial nerve ending receptors with the dermal meridians. METHODS: The experiment was conducted in the Biomedical Laboratory for Tumors of the Lianyungang First People's Hospital between October and November 2004. Reverse neural trace was performed by gastric adventitial injection of fluorescent riboflavin(a kind of neural tracers) in the gastric corpus, gastric fundus, cardia and pylorus of 4 rabbits. After the rabbits survived 18 to 24 hours and 30 to 36 hours, frozen serial sections were made in C2 to L5 spinal ganglia, cervical and thoracic sympathetic ganglia, inferior mesenteric ganglion, cerebrum, cerebellum, thalamus, brain stem, spinal cord, stomach, small intestine, myocardium, lung, sacral spinal muscle in sacrum, liver, urinary bladder, spleen, abdomial aorta, inferior mesenteric artery, abdominal lymph node, dermis in the involved region, dermis and ear in sacrum respectively. The sections were made into 2 series: one was stuck in the distilled water, and then observed under an Olympus ZX-004 fluorescence microscope directly, and the other was stained with hematoxylin and eosin, and then observed under the fluorescence microscope to determine whether they were associated with the pathway of intracutaneous receptors.RESULTS: All the rabbits were sampled successfully, and 288 pieces of films were gotten. ①At 18 to 24 hours after trace, labeled cells were found in the C3 to L3 spinal ganglia, especially more in the T3 to T12 spinal ganglia Labeled cells were also found in cervical and thoracic sympathetic ganglia, and inferior mesenteric ganglion Parasympathetic neurons were labeled below gastric mucous membrane A lot of ganglion cells were labeled on and below small intestinal mucous membrane Dense fluorescence was found in the anterior and posterior horn of spinal cord, sarcolemma of sacral spinal muscle, adventitia and mucous membrane of stomach and urinary bladder walls, myocardial endometria and adventitia, abdomial aorta, adventitia and mucous membrane of mesenteric artery walls, and ansa of intracutaneous blood capillary, however, dense fluorescence was not found in the cerebrum, cerebellum, thalamus and brain stem. ②At 30 to 36 hours after trace, most of the labeled cells in the C3 to T3 spinal ganglia disappeared, especially in the T3 to T10 spinal ganglia Labeled cells in cervical and thoracic sympathetic ganglia, and inferior mesenteric ganglion were similar to that at 18 to 24 hours after trace, and the region of dense fluorescence was the same as that at 18 to 24 hours after trace.CONCLUSION: The pathway net of gastric adventitial nerve ending receptors of rabbits were mainly located in and before the spinal cord, but not associated with high center, and it was inter-overlapped with the pathway net of gastric adventitial nerve ending receptors in the involved region at spinal ganglia, sympathetic ganglia and the neurons of posterior sympathetic ganglia. AIM: To further identify the relationship between pathway of vascular nerve terminal receptor and pathway of cutaneous terminal receptor, and analyze the pathway net of vascular receptor by means of morphological method in rabbits.METHODS: The experiment was conducted in the Biochemical Laboratory of Tumor, First People's Hospital of Lianyugang from October to November 2004. Four female rabbits were selected and divided randomly into retrograde neural tracing 18-24 hours group and 30-36 hours group with two rabbits in each group. ② Rabbits of the two groups were injected with 1 g/L fluorescent riboflavin (Sigma, 74681-68-8) in the vein of ear edges under urethane anesthesia and then received 500 μL retrograde neural tracing. The rabbits in the retrograde neural tracing 18-24 hours group survived 18-24 hours, and rabbits in the retrograde neural tracing 30-36 hours group survived 30-36 hours. ③ The frozen serial sections were made in spinal ganglia from C2 to L5, cervical and thoracic sympathetic ganglia, inferior mesenteric ganglion, cerebrum, cerebellum, thalamus, brain stem, spinal cord, stomach, small intestine, myocardium, lung, kidney, sacral spinal muscle in sacrum, inferior mesenteric artery, dermis in the referral area, dermis and ear in sacrum respectively. The sections were made into 2 series: one was stuck in the distilled water, then observed under an Olympus ZX-004 fluorescence microscope directly to find fluorescence marked neuron and fluorescence dense area of nerve terminal. The other was stained with hematoxylin and eosin, then observed under the fluorescence microscope to determine whether there was new discovery.RESULTS: Four rabbits were involved in the result analysis. Retrograde neural tracing 18-24 hours group: Labeled cells were found in cervical, thoracic sympathetic ganglia and inferior mesenteric ganglion. Parasympathetic neurons were labeled below gastric mucous membrane. A lot of ganglion cells were labeled on and below of small intestinal mucous membrane, while there were no labeled cells in C2 to L5 spinal ganglia, but dense fluorescence were found in outer membrane of ganglia. Labeled cells and fluorescence dense area were not found in the anterior and posterior horn of spinal cord, but dispersed fluorescence was found. There were no labeled cells and fluorescence dense area in cerebrum, cerebellum, hypothalamus, whereas significant fluorescence dense area were found in sarcolemma of sacral spinal muscle, tracheal intima and adventitia, myocardial intima and adventitia, renal glomerulus and ansa of intracutaneoous blood capillary. Dense fluorescence was in membrane of mesenteric artery walls. After hematoxylin and eosin staining, there were labeled cells in inferior mesenteric ganglion, nucleus, which were golden, and the cytoplasm was blue and black. Parasympathetic neurons were labeled below gastric mucous membrane. ② Retrograde neural tracing 30-36 hours group: there were no labeled cells in C2 to L5 spinal ganglia, and labeled cells in cervical, thoracic sympathetic ganglia and inferior mesenteric ganglion were similar to those in the retrograde neural tracing 18-24 hours group, and it was the same in the region of fluorescence dense area.CONCLUSION: The pathway net of vascular nerve terminal receptor in mainly composed of sympathetic ganglion, postganglionic fiber and parasympathetic nerve, the ascending pathway net can reach to spinal ganglia and spine, and the descending pathway net can reach to sarcolemma of sacral spinal muscle, tracheal intima and adventitia, myocardial intima and adventitia, renal glomerulus, ansa of intracutaneous blood capillary and artery walls. The nter-overlapped with the pathway net of cutaneous terminal receptor in the gastric referral area at sympathetic ganglion, postganglionic fiber were involved in the ascending and descending pathway net.
