Microcalorimetry was used to determine the actions of some schiff base metal complexes on the Aerobacter aerogenes. Difference in their capacities to inhibite the metabolism of this bacterium, as judged from the rate constant k (in log phase), varied with the different schiff base metal complexes. The growth rate constants of the bacteria in the presence of Co( II ) - NG(NG: D - glucosamine - β - naphthol aldehyde) decreased with the increasing of its concentrations , but the relationship between k and c is...

Microcalorimetry was used to determine the actions of some schiff base metal complexes on the Aerobacter aerogenes. Difference in their capacities to inhibite the metabolism of this bacterium, as judged from the rate constant k (in log phase), varied with the different schiff base metal complexes. The growth rate constants of the bacteria in the presence of Co( II ) - NG(NG: D - glucosamine - β - naphthol aldehyde) decreased with the increasing of its concentrations , but the relationship between k and c is not of good linearity . In the case of Fe( II )- NG the rate constants keep unvaried with the variation of its concentrations. It seems no rule in the inhibitory effect of Zn( II ) - NG,Cu( II ) - NG on Aerobacter aerogenes. We also found that the relation between k and Pmax (the maximal thermal power) is a good linear equation, so the k can be replaced by Pmax at the same experimental conditions.

采用微量热法测定了产气杆菌在四种Schiff碱金属配合物作用下的代谢热谱,以其对数生长期的生长速率常数k表征了Schiff碱金属配合物对其细菌生长的抑制. 不同的配合物其抑制情况不同.Co(Ⅱ)-NG(NG:D-氨基葡萄糖-β-萘酚醛Schiff碱)对产气杆菌的抑制作用最大,随浓度c的增加抑制作用增大,在350μg·mL~(-1)时可完全抑制其生长,但k-c并不呈线性关系;Fe(Ⅱ)-NG对产气杆菌的抑制较小,且在50~300μg·mL~(-1)浓度范围内k与浓度无关,在350μg·mL~(-1)时才略表现有一定的抑制作用;Cu(Ⅱ)-NG和Zn(Ⅱ)-NG对产气杆菌的抑制作用也较小,且k随浓度的变化不规则.同时发现热谱上最大热输出功率P_(max)与生长速率常数存在正比关系,进而推出在实验条件相同的情况下可用P_(max)代替k.

Results:(1)Level of NAG,γ-GT,LDH-L and ALP were significantly higher in altitude subjects compared with sea level,( P <0 05 or 0.01),(2)level of NAG and ALP at altitude of 5 380m were higher than at 3 700m( P <0 01),but there was no remarkably difference between γ-GT and LDH-L,(3)after 6 months acclimazation level of NAG at 5 380m altitude was higherthan at 3 700m(P>0.05),and level of γ-GT,LDH-L and ALP had not a significantly difference( P >0 05),(4)level of ANG and r-GT on 7th exposure day at 3 700m altitude were higher than in sea level ( P <0 05) but no a difference between LDH-L and ALP( P >0 05),(5)NAG,γ-GT,LDL-L and ALP level on 7th day acclimazation of 5 380m were significantly higher than after 6 months acclimazation (P <0 05 or<0 01).Conclusion:The cellular penetration of renal tubular epithelium and glomerular filtration rate increase at high aititude migrants,and reabsorption of renal tubules is decrease.

目的:探讨高原低氧移居者尿液酶N—乙酰—β—D—氨基葡萄糖甘酶(NAG),γ—谷氨酰转移酶(γ—GT)、乳酸脱氢酶-L(LDH—L)、碱性磷酸酶(ALP)的排泄变化。方法:对从平原进驻海拨3700m和5380m高原第7天和半年的某部官兵进行尿液检测,且与平原健康青年作对照。结果:高原低氧环境下尿液酶NAG、γ—GT、LDH—L和ALP活性均明显高于平原(P<0.05或P<0.01);初入高原第7天,海拔5380m较3700mNAG和ALP活性增高非常显著(P<0.01),γ—GT和LDH—L活性无显著性差异(P>0.05);高原居住半年后,NAG活性5380m较3700m增高显著(P<0.05),γ—GT、ALP和LDH—L活性变化无显著性差异(P>0.05)。进驻3700m第7天较半年时NAG和γ—GT显著增高(P<0.05),LDH—L、ALP变化无显著性差异(P>0.05);5380m第7天较半年时NAG、γ—GT、LDH—L和ALP均显著增高(P<0.05或P<0.01)。结论:高原低氧可导致肾小管上皮细胞通透性增强,重吸收功能降低,肾小球滤过率增加。

The β-N-Acetyl-D-glucosaminidase (NAGase,EC 3.2.1.30) can catalyzes the cleavage of N-acetylglucosamine polymers.It is one of the chitinases and cooperates with endo-chitinase and exo-chitinase to disintegrate chitin into N-acetylglucosamine.The NAGase was purified from the viscera of Turbo cornutus Solander by ammonium sulfate fractionation and gel filtration column chromatography on Sephadex G-200 and twice ion exchange column chromatography on DEAE-Sephadex A-50.The purified enzyme preparation was homogeneous as judged by FPLC and polyacrylamide gel electrophoresis.The specific activity of the enzyme was 1 448 U·mg~(-1).The characteristic peak of UV-absorption spectrum of the enzyme was found at 275 nm and that of the intrinsic fluorescence emission was at 340 nm.The optimum pH was determined to be at pH 4.5 and the optimum temperature was at 45℃.The enzyme was stable in the pH ranges from 3.5 to 6.0,the enzyme activity rapidly decreased while the pH value was lower than 3.5 or higher than 6.0.The enzyme has good thermal stability under 40℃.The enzyme follows typical Michaelis-Menten kinetics for the hydrolysis of pNP-β-D-GlcNAc in 0.1 mol·L~(-1)Citric-Na_2HPO_4 buffer at 37℃.The K_m and V_m values were determined to be 3.13 mmol·L~(-1) and 17.68 μmol·L~(-1)·min~(-1),respectively by plot of Lineweaver-Burk.

以蝾螺内脏为材料,通过硫酸铵沉淀分级分离、SephadexG 200分子筛柱层析和两次DEAE SephadexA 50离子交换柱层析纯化,获得聚丙烯酰胺凝胶电泳单一纯的N 乙酰 β D 氨基葡萄糖苷酶酶制剂.纯酶的比活力为1448U·mg-1.酶的紫外特征吸收峰在275nm处,内源荧光发射峰在340nm处.以对 硝基苯 N 乙酰 β D 氨基葡萄糖为底物,研究酶催化底物水解的反应动力学,结果表明:酶的最适pH为4.5,最适温度为45℃.该酶在pH3.5~6.0区域较稳定,而在pH>7能很快失活;在40℃以下处理30min,酶活力保持稳定,高于40℃,酶稳定性较差,很快失活.酶促反应动力学符合米氏双曲线方程,测得米氏常数Km为3.13mmol·L-1,最大反应速度Vm为17.68μmol·L-1·min-1.
