With acute renal damage induced by a single subcutaneously administered cadmium-metallothionein(CdMT)in male Wistar rats was used as a modle for chronic Cd poisoning, the experiment was performed to search the effects of lipid peroxidation(LPO)on renal damage of CdMT. The morphologic changes in renals of intoxixicated rats were found as a widerspread vesiculation of proximal tubular cells with mitochondrial alterations and choronic retrograde degeneration,and urinary biochemical indicators for renal tubular...

With acute renal damage induced by a single subcutaneously administered cadmium-metallothionein(CdMT)in male Wistar rats was used as a modle for chronic Cd poisoning, the experiment was performed to search the effects of lipid peroxidation(LPO)on renal damage of CdMT. The morphologic changes in renals of intoxixicated rats were found as a widerspread vesiculation of proximal tubular cells with mitochondrial alterations and choronic retrograde degeneration,and urinary biochemical indicators for renal tubular injury and dysfunction were determined in the animals. The levels that renal glutathione (GSH) was significantly decreased and malondialdehyde (MDA) increased,which keep well correlationships with that of rGT and NAG in urine of rats injected with CdMT, suggesting that LPO is partly involved in the tubular damage induced by CdMT.

Peritoneal macrophages of mice were isolated and cultured in vitro,peri-toneal macrophages fluid(5×10 ̄5/ml)were incubated in 24-well tissue culture plates.1mlof the fluid were added into every well.50μl,100μl,200μl of polydatin(0.5%)wereadded respectively into these wells before and after endotoxin-stimulation. Concentrationsof TNF and NAG in the supernate were estimated.Result showed polydatin decreasedthe level of TNF and NAG in the supernate of endotoxin-stimulated macrophage.

收集小鼠腹腔巨噬细胞,调整细胞浓度为5×10 ̄5/ml,加入24孔培养板每孔1ml,内毒素刺激前后不同时间加入白藜芦醇甙培养,检测上清中肿瘤坏死因子,溶酶体酶(N-乙酰-β-D氨基葡萄糖苷酶NAG)的变化,结果说明:白藜芦醇甙可减少上清中TNF_α和NAG的量,通过此可减少内毒素刺激产生的由TNF_α和NAG介导的组织损伤。
