In this paper, we report the experimental treatment of CCl4 induced hepatic fibrosis of rats with hepatocyte stimulating substance (HSS), as well as their hydroxyproline trend. The results revealed that the hepatic hydroxyproline contents of the HSS-treated and HSS-prevented animals were significantly less than those of the control group (P<0.01). In this paper, we report the experimental treatment of CCl4 induced hepatic fibrosis of rats with hepatocyte stimulating substance (HSS), as well as their hydroxyproline trend. The results revealed that the hepatic hydroxyproline contents of the HSS-treated and HSS-prevented animals were significantly less than those of the control group (P<0.01). The serum albumin levels of HSS prevented animals exceeded other groups (P<0.05). Hepatocyte degeneration and fibrous tissue hyperplasia in HSS-treated and HSS-prevented group were significantly less than control (P<0.05). Blood ALT levels showed no statistically valid diference among three groups. This suggested that HSS inhibited hepatic fibrosis and improved hepatic systhesis function. This effect might be due to the stimulating action of HSS particu-lary on damaged liver cells, promoting regeneration, and selectively increased the resolution of collagen.

用新生小牛肝中提取的肝细胞刺激物质(HSS)处理CCl_4实验性肝纤维化的大鼠。结果发现HSS预防组和治疗组的肝组织羟脯氨酸含量明显低于病理对照组(P<0.01),血清白蛋白水平,HSS预防组高于其它各组(P<0.05),肝组织内的变性坏死及纤维组织增生,HSS预防组及治疗组也较病理对照组为轻(P<0.05)。说明HSS具有减缓肝纤维化,提高血清白蛋白的作用,可能与HSS刺激肝细胞再生、选择性增加胶原降解有关。

Therefore,we concluded that HSS is a potential factor which can stimulate liver regeneration. Perhaps,it can be used in clinic. It may be better than the therapy of Insulin plus Glucagon which is widely used in clinic recently.

本研究采用体外细胞培养分组对比法检测肝再生刺激因子(HSS)、胰岛素或/和胰高血糖素对大鼠原代培养肝细胞和人肝肿瘤细胞株HepG_2 DNA合成(肝细胞再生)的作用。结果发现,HSS无论对大鼠肝细胞或人肝肿瘤细胞均具有较强的刺激DNA合成的作用(与对照组相比分别增加1.14和1.05倍,P<0.01)。而对人纤维母细胞则无促进DNA合成的作用。低浓度胰岛素或/和胰高血糖素对肝细胞DNA合成无作用。高浓度胰岛素(10~(-5)M)具有中等度促进肝再生的作用(与对照组相比增加40%,P<0.05)。两者联合应用(G+I)似无协同作用。据此,我们认为,HSS是一种潜在的促进肝再生的有效因素,可能具有临床实用价值。体外应用优于现在临床广泛应用的胰岛素加胰高血糖素(G+I)疗法。

本文用大鼠从体内、外观察了前列腺素E_2(PGE_2)对肝再生的作用。重点集中在肝部分切除后所分离的离体枯否细胞(KC)在大肠杆菌脂多糖(LPS)的刺激下合成及分泌PGE_2的能力。结果显示,给大鼠体内注射消炎痛(5mg/kg)抑制KC的PGE_2产生,从而显著地阻碍了肝脏的再生。其DNA合成率明显低于未使用组(P<0.01)。而在DNA合成高峰期(术后24h)所分离的再生肝之KC,其合成PGE_2的能力明显强于假手术组对等数量的KC(PGE_2浓度分别为15ng/ml及6.5ng/ml,P<0.01)。据此我们得出结论,KC通过其内源性PGE_2的产生从而启动肝细胞的再生。
