Endothelial viability and ultrastructural integrity was maintained in most of the corneas even in this one up to 138 hours.Six intact eyes stored in such method were used for penetrating keratoplasty.These grafts were clear at the time of surgery and remained so post-operatively.The electron microscope was used to compare corneas during 46-118 hours of storage in M-K medium of 11 excised corneas,and did not find difference between two methods.Nine of the 11 excised corneas were used for penetrating kerato- plasty.There were clear grafts in 7 cases and there were no primary graft failures.The electron microscope was used to compare these whole eyes stored in M-K medium and the fresh cornea and eye in moist chamber for 118.5 hours.The eye stored in moist chamber showed more extensive deterioration of endothelial cell ultrastructure than the corneas and eyes stored in M-K medium.This studies indicate intact eyes with air injection into the anterior chamber to isolate the endothelium from the aqueous and stored in M-K medium the endothelial cell ultrastructure was as good as that of the excised corneas stored in M-K medium.

ObjectiveTo examine apoptosis in mid term storaged corneas,so as to provide theoretical basis for improving the method of corneal storage.Methods Rabbit and human corneas were stored in M K liquid,M K liquid with epidermal growth factor(EGF)(10?ng/ml)and optisol liquid.Morphological study was performed by light microscope and electron microscope on the occurrence of apoptosis at 3,5,7 d after the corneas were preserved in three kinds of liquids.Quantitative study was also performed on the occurrence of apoptosis at different time by TUNEL and light microscope.Results The characteristic apoptotic cells were observed by light microscope and electron microscope in corneal epithelium,stroma,and endothelium when corneas were preserved in three kinds of liquids after 3,5,7?d.TUNEL positive cells increased evidently when prolonged the time of corneal storage in three kinds of liquids ( P <0 01).TUNEL positive cells of corneas had no differences between preservation in M K liquid and in M K liquid with EGF( P >0 05),but TUNEL positive cells were significantly higher than that of preservation in optisol liquid ( P <0 01).Apoptotic cells were also observed in human corneas that preserved in M K and optisol liquid for 4?d.ConclusionsThe results indicate that apoptosis may be another mechanism of cell death in corneal storage and apoptosis that influences the quality of mid term corneal storage.M K liquid with added EGF has no significant differences in preventing the apoptosis of cornea in mid term liquid.

目的 探讨角膜中期保存过程中细胞凋亡现象,为改进角膜保存方法提供理论依据。方法 采用MK液、MK+EGF液和Optisol3种保存液保存兔和人角膜,用光镜、电镜及TUNEL标记技术检测细胞凋亡发生情况。结果 3种保存液保存3、5、7天的兔角膜,在上皮、基质及内皮细胞层均可见典型的细胞凋亡发生。保存时间不同,保存液不同,TUNEL阳性细胞数不同。MK液和Optisol液保存4天的人角膜同样可见细胞凋亡发生。结论 细胞凋亡是角膜中期保存过程中细胞丧失的另外一种可能机制;细胞凋亡的发生对角膜中期保存效果存在一定的影响;表皮生长因子在角膜中期保存过程中没有阻止角膜细胞凋亡发生的作用。
