Objective To explore the effects of the new gasoline additive, methyl tertiary butyl ether (MTBE) on cell cycle and cell apoptosis. Methods Flow cytometry was used to evaluate the effect of MTBE (1, 2, 4 μl/ml, 24 h) on NIH/3T3 cell cycles; and the effect of MTBE on Hela cell apoptosis was evaluated by detecting cell survival using crystal violet staining. Results Flow cytometry showed that MTBE could change NIH/3T3 cell cycles, decrease the number of cells in S stage, and arrest cells at G 2+M stage. The results suggested that MTBE could affect NIH/3T3 cell cycles and induce cell proliferation. This situation existed 48 hours after the treatment, and cell cycles came back normal 96 hours after the treatment. By detecting cell survival using crystal violet staining, we found that MTBE could inhibit the apoptosis of Hela cells which was induced by tumor necrosis factor (TNF)α and cycloheximide. Conclusion MTBE′s carcinogenicity to animals may relate to induction of cell proliferation and inhibition of cell apoptosis.

目的 探讨汽油添加剂甲基叔丁基醚 (MTBE)对细胞周期和细胞凋亡的影响。方法采用流式细胞仪检测MTBE对NIH/3T3细胞周期 (染毒浓度分别为 1、2、4μl/ml,染毒时间为 2 4h)的影响 ;应用结晶紫染色法 ,通过检测Hela细胞的存活情况 ,了解MTBE对细胞凋亡的影响。结果 流式细胞仪结果显示 ,MTBE可使NIH/3T3细胞周期发生改变。表现为S期细胞减少 ,而处于G2 +M期的细胞增多 ,提示MTBE可能具有影响NIH/3T3细胞周期的作用 ,可诱导细胞增殖。这种作用持续存在于染毒后 48h的细胞中 ,到染毒后 96h ,细胞周期基本恢复正常。结晶紫染色法通过检测Hela细胞的存活情况 ,发现MTBE可明显拮抗肿瘤坏死因子 (TNF)α和放线菌酮所诱导的Hela细胞的凋亡。结论 MTBE可诱导细胞增殖和抑制细胞凋亡 ,这是其动物致癌性的可能机制之一。

Methyl tertiary butyl ether(MTBE) is a new gasoline additive used to increase gasoline octane value and decrease the exhaust of some harmful pollutants,but there is still debate on its toxicology. With the expansive use of MTBE in China,it is imperative to explore homemade MTBE toxicology. In this artcle,subchronic oral MTBE assay was carried out on Sprague Dawley rats,the doses were 1 000 mg/kg,600 mg/kg,200 mg/kg and 0 mg/kg body weight,once daily,five days weekly,for 13 weeks. The serum biochemical indexes were detected,meanwhile the damage of ultra structure of nephric cortex cells was observed with electron microscopy. The biochemical index including LDH,AST,ALT,TP,A/G,BUN and Cr were analyzed,the results showed that there was no significant difference between test and control groups except a distinct decrease of LDH and AST,but they are within the normal level. The ultra structure was characterized by appearance of primary lysosomes,proliferation of mitochondrion as well as swell of podocyte and base membrane. [Conclusion] The damage is still in the degree of compensation,and no obvious difference of serum enzymes and proteins is observed. There was no appearance of damage on the function of kidney.

[目的 ]甲基叔丁基醚 (MTBE)是一种新型的汽油添加剂 ,常用来提高汽油辛烷值和减少汽车尾气中有害物质的排放。它的毒性研究国内外学者的结论尚不一致。随着国内MTBE的使用推广 ,有必要了解国产MTBE的毒性。[方法 ]SD大鼠经灌胃给MTBE 10 0 0、6 0 0、2 0 0mg kg ,每天一次 ,每周 5d ,共 13周亚慢性染毒。采用生化分析仪分析了大鼠相关血清生化指标的改变。并用电镜观察了大鼠肾皮质区超微结构的变化。 [结果 ]血清生化分析结果表明各剂量组动物乳酸脱氢酶 (LDH)和谷草转氨酶 (AST)有明显下降 ,但仍在正常范围内。其他指标如谷丙转氨酶 (ALT)、血清总蛋白 (TP)、白球蛋白比值 (A G)、尿素氮 (BUN)、肌酐 (Cr)未见明显改变。电镜观察到肾小管细胞胞浆内有初级溶酶体、线粒体异常增殖 ,10 0 0mg kg高剂量组中肾小球系膜基质与足细胞足突肿胀现象。 [结论 ]肾脏细胞对MTBE的毒作用处于一种代偿状态 ,血清生化指标未见改变 ,尚不能认为MTBE对动物肾功能有明显损伤作用

Objective To assess the genetic toxicity of methyl tertiary-butyl ether (MTBE), an unleaded gasoline additive.Methods 40 mice were distributed in four groups randomly. The change of the marrow micronucleus in polychromatic erythrocytes (PCE) and semina deformity ratio were detected. Results The marrow micronucleus in PCE and semina deformity ratio in MTBE exposed groups were significantly increased, especially in 1 600 mg/kg-bw group ( P < 0.05) . The forms of malformed sperm were non-hook, small head, un-shaped and folded in tail. Conclusion MTBE might have the potential genetic toxicity in mice.

目的 了解汽油添加剂甲基叔丁基醚(MTBE)的遗传毒性。方法40只健康昆明种成年小鼠,随机分为4组。用甲基叔丁基醚进行灌胃染毒,染毒剂量分别为1 600、400、100和0 me,/kg体重。每天一次,每周5 d,共计40 d,观察其对小鼠骨髓嗜多染红细胞微核率和精子畸形率的影响。结果 MTBE染毒各组体重轻度降低,染毒组随剂量增加骨髓嗜多染红细胞微核率及精子畸形率增高,高剂量组与对照组比较差异有显著性(P<0.05);畸形精子形态主要表现为无钩、小头、无定型及尾折叠等改变。结论MTBE对昆明种小鼠具有一定的遗传毒性,能使小鼠骨髓嗜多染红细胞微核率、精子畸形率增高。
