The result of acomparative study demonsats tha the strongly hydrophobic residues at position 18 and 17 in TypesⅠ and Ⅱ neurotoxins, respechvely, play a critical role in high affinity channel binding, whilehydropobic residues Ⅱe-43 and Ⅱe-40 in both Types Ⅰ and Ⅱ neurotoxins are probably an importantstructural detendnanL the Ap-A and Ap-B residues at positions 12 and 49 are essential for bothhigh affinity and isoform discrimination. Consequentiy, thsese characteristics are essenhal for theapplication of the anemone toxin to the design of new cardiotonic drugs.

海葵神经毒素是一类能与可激动细胞电压门控钠通道发生亲和作用的海洋多肽毒素。根据80-90年代中期国际上对海葵神经毒素的研究成果,重点对其结构特征、结构与功能的关系进行综合评述。已有的结果表明,所有已发现的海葵神经毒素其同源性较高,拥有相似的二级结构和三维结构;不规则环状结构的正确构象对毒素的心肌刺激活性是至关重要的。通过比较研究方法,作者认为:typeⅠ毒素的残基18和typeⅡ毒素的残基17侧链具有强流水性,对毒素的活性起关键作用;而typeⅠ毒素残基43和typeⅡ毒素残基40的疏水烷基侧链,可能对海葵毒素的形成起决定作用。

Recombinant salmon calcitonin hexa polymer , salmon calcitonin octo polymer and human prourokinase had more soluble form in cytoplasm of GJ980 mutants than in wild type strain, while 3D protein, which has no cysteine residue, still remain in insoluble form. Results indicate the GJ980( trxB - ) strain allowed the formation of disulphide bonds in the cell cytoplasm which is believed to encourage correct folding and soluble expression of the recombinant proteins.

以人工设计的、不含半胱氨酸残基的三元蛋白 ,六聚和八聚鲑鱼降钙素融合蛋白和人尿激酶原等具有不同半胱氨酸残基含量的外源蛋白质为例 ,利用大肠杆菌硫氧还蛋白还原酶基因缺陷菌株GJ980 (DE3trxB-) ,探索把以包涵体形式表达的外源蛋白质变为可溶性表达的可能性及其规律。研究表明 :由于硫氧还蛋白还原酶基因的缺陷所引起的细胞质氧化还原态势的变化 ,使一些在普通大肠杆菌宿主中以包涵体形式表达 ,含有半胱氨酸残基的重组蛋白 ,在GJ980中能在一定程度上以可溶性蛋白质形式表达 ;不含有半胱氨酸残基的重组蛋白在GJ980中仍以包涵体形式表达。推测重组蛋白在GJ980细胞质中形成二硫键对其正确构象的形成具有一定的作用
