During the megasporogenesis of rice(Oryze sativa), nucellar cells retain more characteristics of meristematic cells. They are regular in form, highly cytoplasmic and rich in ribosomes, either suspended in the cytoplasm or attached to the endoplasmic reticulum and on the outer nuclear membrane. Reaction product of acid phosphatase is localized primarily in the small vacuoles, and occasionally in the dictyosome cisternae and vesicles of the golgi body. With the differentiation of embryo sac, a series of complicated changes occur in the inner layers of the nucellus. Vacuoles encapsulate, frequently and continuously, portions of cytoplasm containing ribosome, plastids, mitochondria and vesicles, resulting in the formation of larger vacuoles with large amount of reaction product of acid phosphatase. The further enlargement ansubsequent rupture of the large vacuoles cause the destruction of the nucellar cells. The second change include the dilation of rough endoplasmic reticulum cisternae and their engulfing and sequestering cytoplasmic components, and the occurrence of the membrance-bound contents within the nuclear envelope. The third change was the seperation of the plasmalemma from the cell wall and the subsequent formation of the large space between the plasmallemma and the cell wall, in which numerous small vacuoles and heavy reaction product of acid phosphatase were observed. Finally protoplasms of the nucellar cells shrinks dramatically and convert to disorganized electron-dense debris.
