We studied this intranuclear body,called mininucleolus in our paper,and found that the body appeared as an ovoid or spherical structure about 0.3 to 0.5 μm in diameter.The mininu- cleolus was shown,with conventional TEM staining,to be the same electron-dense as the con- densed chromatin,but no structural connections could be found between them.The results of Bernhard's staining and the protein specific staining indicated that the mininucleolus contained both RNA and proteins which formed ribonucleoprotein(RNP)particles about 15 μm in diame- ter.These particles are very similar,in their ultrastructure and chemical composition,to the granular comoonents of the nucleolus.A careful examination revealed that the mininucleoli were formed in the granular region of the nucleolus.After they became separated from the nucleolus,these bodies were distributed throughout the nucleoplasm,suggesting that they may move from the nucleolus to the nuclear membrane.Based on these data,we suggest that the mininucleolus is a non-chromatin intranuclear structure which is formed in the nucleolus and may have a function of transporting ribosomal RNA from the nucleolus to the cytoplasm.

以蚕豆(Vicia faba)根端分生组织细胞为材料研究了微核仁的超微结构和细胞化学特点。结果表明;微核仁是直径0.3—0.5μm 的卵圆形或球形结构。常规染色时,微核仁与集缩染色质的电子密度相仿,但两者之间在结构上没有任何联系。细胞化学研究指出,微核仁含有 RNA 和蛋白质,其结构成分主要是与核仁颗粒组分十分相似的 RNP 颗粒。报道了植物细胞核中微核仁发生于核仁的过程并对微核仁的本质和功能进行了讨论。

With alkaline elution assay, effects of domestic carminomycin on DNA single-strand breaks were observed in leukemia P388 cells. The effects were dose-dependent, timedepen dent and irreversible. Under electron microscope, it was obser/ced that carmino mycin caused a nucleolar "Cap" forming and a nucleolar segregation or fragmentation.

用碱洗脱技术观察了国产洋红霉素(CMN)引起小鼠淋巴白血病P_(368)细胞单链DNA的断裂作用。在CMN0.25—2.0μg/ml范围内,这种作用是剂量依赖性的。电镜观察研究CMN对P_(388)细胞核仁超微结构的影响。CMN 1.0μg/ml作用2小时,核仁空泡增多、微球体出现;作用4小时;核仁“帽”形成;作用8小时,部分细胞核仁成分碎裂。

Kunming mouse oocytes (17 h after hCG injection) were treated with the condition of 1.0 kV/cm in strength, 320 μs in duration and 3 pulses at intervals of 1 second, and ultrastructures of mouse oocytes before and after electrical activation were observed. The results showed that in most mouse oocytes, the ultrastructures of cell envelop, organelle and nucleus appeared gradually activated 1 7 h after the treatment of electrical activation, and those of mitochondrion and nucleolus in a few mouse oocytes appeared...

The results showed that in most mouse oocytes, the ultrastructures of cell envelop, organelle and nucleus appeared gradually activated 1 7 h after the treatment of electrical activation, and those of mitochondrion and nucleolus in a few mouse oocytes appeared abnormally. The ultrastructural observations indicated that the above mentioned condition of eletrical activation can efficiently activate most mouse oocytes, and a few mouse oocytes treated are abnormal.

为了给小鼠卵母细胞电活化的深入研究提供形态学依据,以1.0kV/cm、320μs、3次直流电脉冲(每次间隔1s)的条件,对小鼠卵母细胞(hCG后17h)进行了电活化处理,对活化前后小鼠卵母细胞的超微结构进行了观察。结果显示,电活化刺激后1~7h,大多数小鼠卵母细胞的细胞膜、细胞器和细胞核的超微结构呈现出功能活动逐渐活化的结构相,而少数卵母细胞中泡状线粒体和核仁的超微结构见有异常变化。结果表明,本试验所采用的电活化条件可有效地使小鼠卵母细胞发生活化,而少数卵母细胞中泡状线粒体和核仁出现异常,说明并非所有的活化卵都是正常的。
