bjectives To explore the probability that defected peripheral nerve was repaired with the composite of muscle cells and Schwann cells.Methods The composite of autogenous muscle fragment and autogenous nerve fragment were wrapped up with fascia to repair sciatic nerve defect of 15mm in rats.The nerve condution velocity,function recovery,histological observation were examined after 8 weeks of operation.Results The composite filled with muscle fragment and nerve fragment were similar to those filled with nerve fragment of same amount 15mm sciautic nerve,better than that filled with 15mm muscle fragment or 7.5mm sciautic nerve fragment,and close to that implanted with 15mm autogenous nerve.Conclusions It was thougt that the composite of the muscle cells could act as scaffold for Schwann cells attachment and the small amount of Schwann cells played a role in guiding regenerated nerve growing through the defect.

Objective Lack of transplant material is a main problem in the repair and reconstruc- tion of tissue or organ defects.To seek the succedaneum of transplants,we studied and used tissue-engi- neered growth hyaline cartilage.Methods The articular hyaline cartilage obtained from newborn calf within 6 hours of sacrifice was enzymatically dissolved.The cbondrocytes were then seeded onto the medi- cal collagen membrane of guided tissue regeneration in a 24 well plate in RPMI1640 medium.One week later,the chondrocyte medical collagen membrane of guided tissue regeneration complex was implanted subcutaneously into the back of nude mice.Eight weeks after implantation,the animals were sacrificed. Results The present study seems to demonstrate that the cartilage-like tissue was strong enough to be transferred after being implanted for 8 weeks.The cartilage-like tissue was proved to be tissue-engineered hyaline cartilage by HE stain.The chondrocytes could secrete chondroitin sulfate as proved by Lev-Spicer stain.Conclusion Tissue engineering technique was used to make tissue-engineered cartilage in vitro.It will provide a new transplant material for the repair of tissue or organ defects.

目的解决修复重建组织器官缺损所需移植材料的问题,利用组织工程技术,进行组织工程牛透明软骨的实验研究。方法用分离获得的牛软骨细胞,与医用组织引导再生胶原膜体外联合培养1周后,以软骨细胞可降解材料复合物的形式植入裸鼠体内,8周后对所获得的组织工程软骨样组织进行形态学研究。结果在实验获得的组织工程软骨样组织中有软骨细胞的存在,并且可以分泌硫酸软骨素。证实为组织工程牛透明软骨。结论实验结果表明应用组织工程技术,在体外进行组织工程软骨的制作,可能成为解决整形外科领域修复缺损用移植材料不足的方法之一。

目的 了解组织工程研究的基本原理 ,完善制作组织工程化人软骨的技术方法 ,探讨天然可吸收材料组织 ,引导再生胶原膜作为人软骨细胞体外培养支架的可行性 ,为广泛开展组织工程的研究与应用开辟新的途径。方法 利用组织工程技术制作人组织工程化软骨 ,并应用组织学方法 ,对获得的人组织工程化组织形态和功能进行表征。结果 培养的软骨细胞均匀沉降于医用组织引导再生胶原膜上 ,1周后形成一层乳白色软骨样组织 ;植入 8周后从裸鼠体内获得软骨样组织 ,组织学检测证实软骨细胞的存在 ,软骨细胞可以分泌硫酸软骨素。结论 医用组织引导再生胶原膜以其特有的三维结构和细胞网结功能 ,可以作为组织工程技术应用研究中软骨细胞培养的支架 ;利用组织工程技术 ,能够完成人组织工程化软骨的制作。
