To investigate further the possible molecular mechanism of IL-6 on mesangial cell proliferation, we examined the effect of IL-6 on c-myc protooncogene mRNA expression in cultured human mesangial cells (MC)by dot blot hybridization. Mesangial cells(2 x10~6)made quiescent by exposed to a defined serum-free medium were used in the experiment. IL-6 and Emodin were added ndependently or together to the media of cultured human MC at zero time. It was found that the c-myc mRNA expressed was enhanced markedly in MC treated with IL-6. This effect was dose-dependent and blocked by Emodin. In conclusion: (1) In quiescent MC, a low basal xpression of c-myc mRNA could be detected. (2) Exposure of MC to IL-6 resulted in a marked increasement of c-myc mRNA expression in adose-dependent manner. This sequential activation of protooncogene and cellular reaction may be an important mechanism in IL-6 induced MC proliferation. (3) Emodin has down-regulating effect on c-myc mRNA overexpression of MC induced by IL-6. It is speculated that Emodin might inhibit MCproliferation through this path way.

本文应用体外细胞培养和斑点杂交技术,首次对重组人IL-6对人系膜细胞c-myc原癌基因mRNA的表达以及大黄素对IL-6的作用进行了观察。发现IL-6能够促进人系膜细胞c-myc原癌基因mRNA的增殖表达,该作用呈明显的剂量依赖性,而大黄素能够拮抗IL-6的上述作用。本项研究阐明了IL-6促进系膜细胞的可能机理,为进一步论证大黄素对系膜细胞功能的影响,大黄素与细胞因子间的相互作用以及大黄在防治肾小球硬化的作用奠定了基础。

Increased mesangial cell

本文以纤维连接蛋白(FN)为指标,观察了大黄组份之一大黄素素,对培养人系膜细胞细胞外基质产生的影响、结果用ELISA检测、免疫荧光染色以及系膜细胞小丘形成的大小、多寡等多种指标证明了大黄素能够浓度依赖性地抑制系膜细胞分泌的和细胞膜相关的FN水平,并能拮抗TGFβ对系膜细胞FN产生的刺激作用。大黄素的这一药理作用可能与大黄治疗多种慢性肾脏疾病的机理有关。

目的探讨环孢素A(CsA)和雷公藤单体T4(T4)的免疫抑制机制以及两药联合使用的效应,特别是小剂量CsA、T4联合获得较强免疫抑制效应的可能性。方法用3H-TdR掺入率法测定人系膜细胞(MsC)增殖,用逆转录-聚合酶链反应(RT-PCR)和ELISA法测定细胞IL-6mRNA表达及培养上清IL-6含量。结果CsA和T4均剂量依赖性抑制系膜细胞增殖及IL-6合成,两药联合产生明显的协同作用,小剂量CsA、T4联合与大剂量CsA作用相当。结论CsA和T4具有协同免疫抑制作用
