Objective To investigate the changes of the peripheral blood platelet counting and function the mouse model of idiopathic thrombocytopenic purpura (ITP) and to observe the effect of Zidian Granule (ZG). Methods The peripheral platelet counting was conducted with the automatic hemocyte analyzer; CD41 andCD61 level was measured with flow cytometry. Results The amount of the peripheral blood platelet in ITP model mouse was decreased after injection of antiplatelet serum (APS) and CD41 and CD61 were also decreased.(P < 0.05 as compared with those before injection). ZG in large dose and prednisone both increased the amount of platelet and ZG at large dose could increase the content of CD41 and CD61 to the normal level. Conclusion The platelet counting and function are abnormal in the episode of ITP; ZG can increase the amount of the peripheral blood platelet and restore the aggregation function of platelet, its- term effect is better than prednisone acetate. 

目的观察ITP模型小鼠外周血小板数量与功能的变化情况,探讨血小板在ITP发病中的作用。方法采用全自动血细胞分析仪检测外周血小板计数,采用流式细胞仪检测CD41(抗GPⅢ3a)、CD61(抗GPⅡb)。结果注射抗血小板血清(APS)后ITP模型小鼠外周血小板计数明显下降,同时CD41、CD61也明显降低,差异有显著性意义(P<0.05);紫癜颗粒大剂量组与醋酸泼尼松组均能显著提升血小板计数;紫癜颗粒大剂量组可使CD41、CD61恢复正常水平。结论ITP存在血小板数量的减少及功能的异常;中药复方紫癜颗粒可明显提高外周血小板数量,还可恢复血小板聚集功能,且远期疗效优于醋酸泼尼松。

Method Radioimmunologic techniques were used to detect the levels of IL-2, IL-6 and IL-8. Results As compared with those in the control group, the levels of IL-2, IL-6 and IL-8 were all markedly increased in the peripheral blood of the mouse model of ITP (P<0.05). Conclusion Zidian Granules have marked regulatory effects on the levels of the cytokines in the peripheral blood of the mouse model of ITP. The findings in the animal model suggests that IL-2, IL-6 and IL-8 are related to the occurrence, development and prognosis of ITP.

Objective: To investigate the therapeutic effect of No.2 Zaisheng capsule on idiopathic thrombocytopenic purpura in mice. Methods: The ITP model in mice was made by immunological method. 60 mice were randomly divided into 6 groups,control group,serum control group,model group,positive drug group,low-dose experimental group and high-dose experimental group,10 mice in each group.The mice in experimental group and positive drug group were given different dose drugs by gavage,however,the mice in the rest groups...

Objective: To investigate the therapeutic effect of No.2 Zaisheng capsule on idiopathic thrombocytopenic purpura in mice. Methods: The ITP model in mice was made by immunological method. 60 mice were randomly divided into 6 groups,control group,serum control group,model group,positive drug group,low-dose experimental group and high-dose experimental group,10 mice in each group.The mice in experimental group and positive drug group were given different dose drugs by gavage,however,the mice in the rest groups were given normal saline instead.The general condition of mice during experiment was observed. The mice were killed after 10 days and the mice's body weight, spleen and thymus index, hemogram, myelogram, pathological changes of spleen,T lymphocyte subpopulation were detected. Results: The purpura and gastrointestinal hemorrhage were found in mice of model group, and platelet was significantly decreased, as compared with that of the other groups (P<0.01). The increase ofbonemarrowmegakaryocyte and dysmaturityof marrow cells were found in model group. The experimental group and positive drug group had obvious therapeutic effect,which could increase platelet significantly,as compared with model group(P<0.01).The drugs could relieve obviously dysmaturity of marrow cells,decrease the numbers of bone marrow megakaryocyte,increase significantly the numbers of platelet-produced megakaryocyte (P<0.01),decrease spleen index, reduce the numbers of CD8,as a result,increase the ratio of CD4/CD8. The therapeutic effect was most remarkable in high-dose experimental group. Conclusion No.2 Zaisheng capsule has obvious therapeutic effect on idiopathic thrombocytopenic purpura in mice, which can significantly increase platelet, and its mechanism of action is related to the regulation of immunological function.

目的探讨二号再生胶囊治疗特发性血小板减少性紫癜(ITP)的疗效。方法用免疫法建立小鼠ITP模型,设正常对照组、豚鼠血清对照组、模型组、阳性药组、低剂量实验组和高剂量实验组共6组,实验组和阳性药组灌胃给予不同剂量的药物,其他各组给予等量生理盐水。实验过程中观察小鼠一般情况的改变。给药10d后处死小鼠,检测小鼠体重、脾脏和胸腺指数、血象、骨髓象、脾脏病理和T淋巴细胞亚群等各项指标的改变。结果模型组小鼠出现紫癜和胃肠道出血症状,血小板显著降低,与其他各组相比有显著性差异(P<0.01),并有骨髓巨核细胞数增多,表现出骨髓细胞成熟障碍。实验组和阳性药组对ITP小鼠有明显的治疗作用,表现为血小板明显增多,与模型组相比差异显著(P<0.01),骨髓细胞成熟障碍减轻,巨核细胞数恢复正常,尤其以产板巨核细胞显著增多(P<0.01),脾脏指数降低,外周血CD8降低,CD4CD8比值升高。各实验组中以高剂量实验组效果最为明显。结论二号再生胶囊治疗特发性血小板减少性紫癜模型小鼠疗效显著,尤其以升高血小板的作用最为明显。其作用机制与调节机体的免疫功能有关。
