2. The condition of reverse primer PCR (RP-PCR) and direct primer PCR method in transgenic birch was conformed. The result of reverse primer PCR (RP-PCR) and direct primer PCR associating with Southern blotting could determine the copy number of T-DNA integrated into the host genome.
This paper took the insect-resistant transgenic birch (Betula platyphylla) grown in greenhouse for four years as study materials. The genomic DNA and RNA of leaves were extracted and the integration and the expression characteristic of the extraneous genes in the insect-resistant transgenic birch were analysed by methods of multiplex PCR (MPCR), reverse primer PCR (RP-PCR), inverse PCR (IPCR), RT-PCR, and insect-feeding experiment of the transgenic plants respectively.
The resistances to Lymantria dispar for 39 transgene clones and the control of Betula platyphylla were determined. The result showed that the highest resistant clones with the mortality more than 75% accounted for 15.38%, the middle level resistant clones with mortality of 41%~75% accounted for 46.1%, the lower resistant clones with mortality of 15%~40% accounted for 21.51%, clones with mortality less than 15% accounted for 17.95%.
This report demonstrates that the constitutive expression of this gene in transgenic birch lines increased the resistance of birch against the leaf spot fungus P.
