Antibody-dependent Cell-mediated cytotoxicity (ADCC) of peripheral blood lymphocytes against chicken red blood cells (ChRBC) in the presnece of specific antiserum has been studied in normal subjects and in patients with liver disease. Antibody-dependent Cell-mediated cytotoxicity (ADCC) of peripheral blood lymphocytes against chicken red blood cells (ChRBC) in the presnece of specific antiserum has been studied in normal subjects and in patients with liver disease. The method used in determining ADCC was recommended. ~(51)Cr labelled chicken red blood cells were used as target cells and mixed up with anti- ChRBC serum. Then the isolated peripheral lymphocytes either of normal persons or of hepatic patients as effector cells were added. These specimens were incubated at 37℃ for five hours. The radioactivities of the spcimens were recorded with a scintillation counter. 15 normal subjects and 28 cases of different kinds of liver disease, including 7 cases of advanced primary liver cancers, 9 cases of acute viral hepatitis, 4 cases of chronic persistent hepatitis and 8 cases of chronic active hepatitis, were examined. The results showed that the mean ~(51)Cr net release rate in normal subjects was 59.26±2.86%, while in the patients with liver disease, it was below 31% (p<0.001). The measure-ment of ADCC reaction is a sensitive laboratory method, and it might be useful in assessing the development of the disease and the efficacy of the treatment.

本文报道ADCC的检测方法以及对正常人和肝病患者检测的初步结果。检测方法是以鸡红细胞作靶细胞,人外周血淋巴细胞作效应细胞,采用国产~(51)Cr释放法进行。正常人的~(51)Cr净释放率为59.26±2.86%,晚期肝癌和活动性肝炎(包括急性、慢活肝、慢迁肝)的~(51)Cr净释放率均明显低于正常值,提示ADCC测定对一些疾病的观察有一定临床意义。

The human macrophage cytotoxicity in vitro was observed by inverted microscope with photographical arrangement and time-lapse cinemicrography.This observation revealed dramatic changes in both effectors and target cells. The macrophage cytotoxicity, tumor cell killing, should go through the tumout cell-macrophage membrane contact. The in vitro cytotoxicity of macrophages from patients with stomach cancer, benign stomach diseases and normal adults were studied and different degrees of cytotxicity effect observed....

The macrophage cytotoxicity, tumor cell killing, should go through the tumout cell-macrophage membrane contact. The in vitro cytotoxicity of macrophages from patients with stomach cancer, benign stomach diseases and normal adults were studied and different degrees of cytotxicity effect observed. It was found that pre-cultivation of macrophages from stomach cancer patients still had cytotoxicic effects on gastric carcinoma cell line. In cultivation of macrophages from stomach cancer patients, the cell-free exudate had no influence on the target cells.The experimental results are discussed.

本文应用了附有摄影装置的倒置显微镜与缩时定格显微电影摄影观察了人巨噬细胞体外细胞毒作用。这观察发现了效应细胞与靶细胞之间戏剧性的变化:巨噬细胞细胞毒对肿瘤细胞的杀伤必须通过这两种细胞的膜接触这一步骤。本实验分别观察了胃癌患者、良性胃疾患患者以及正常人的巨噬细胞体外细胞毒作用,发现这些细胞毒作用有强弱程度的不同。实验还发现胃癌患者的巨噬细胞经预培养后对胃癌细胞株仍有细胞毒作用。还看到胃癌患者巨噬细胞培养上清液不影响靶细胞的生长。本文最后对实验观察的结果进行了讨论。
