Autologous B-lymphocytes of Macaca rhesus

用人EBV转化恒河猴的自体B淋巴细胞,再以表达SIV不同抗原成分的重组痘苗病毒VAC-SIVenv、VAC-SIVgag感染,制备51Cr释放试验的靶细胞。恒河猴于制备B淋巴细胞30d后,皮下接种BV-SIVenv亚单位疫苗,每只动物1mL于强化免疫后的30、60、90d,从动物四肢内侧静脉采血,分离淋巴细胞,采用51Cr释放试验检测动物体内的CD8+Tc的动态变化。结果:19只免疫恒河猴中,有3只于强化免疫后呈现明显的CD8+Tc特异性细胞免疫反应,对靶细胞的杀伤活性在16.7%~20.2%之间,井可在体内维持3个月之久;在靶细胞数量不变,渐次增加效应细胞时,免疫细胞的CD8+Tc的杀伤活性随数量增加而增强。从而证明该SIV亚单位疫苗可使部分(16%)免疫动物产生较好的特异性细胞免疫反应。

The method of“fast releasing of LDH”was used in detecting NK activity of PBLs of goats and buffaloes experimentally infected with Fasciola hepatica.The target cells were SP2/0 cells.The results showed that NK activity of goats had two tops,which were suggested to be related with growth of worms or immune state of animals.Close changes of two groups(each infected with 200 or 500 metacercariaes)showed that there was no correlation between NK activity and infective amount.NK activity of infected buffaloes were higher than that of controls' although there was no significant difference.The results also showed that these two kinds of animals had different NK activity levels both in nature and after being infected.NK activity of buffaloes were higher than that of goats,which indicated that buffaloes were more resistant to F.hepatica than goats.

自然杀伤 (NK)活性是在细胞水平反映机体天然免疫力的重要指标之一。本实验以SP2 / 0细胞为靶细胞 (即效应细胞 ) ,采用乳酸脱氢酶(LDH)短程释放法测定人工感染肝片吸虫的水牛和山羊的外周血液中淋巴细胞的NK活性动态变化。从测定结果的相对值来看 ,发现山羊在感染肝片吸虫后NK活性有 2次高峰 ,推测与虫体的发育或感染山羊的免疫状态有关 ;2个感染组 (Ⅰ ,2 0 0个囊蚴 /只 ;Ⅱ ,50 0个囊蚴 /只 )的变化趋势表明 ,感染量与NK活性无相关关系。感染水牛的NK值仅略高于对照组 ,无明显的高峰。而绝对值结果又反映出不同动物的NK活性有差异 ,水牛的NK活性比山羊高 ,说明水牛对肝片吸虫的天然免疫力比山羊强
