随机将174只大鼠分为Ⅰ、Ⅱ、Ⅲ组,每组58只动物。Ⅰ组:用等渗盐水灌胃;Ⅱ组:用毒蕈匀浆液灌胃;Ⅲ组:毒蕈匀浆液灌胃同时腹腔注射山莨菪碱(654-2)注射液,观察肾脏生化和形态学改变。Ⅱ组灌胃后6h肾皮质自由基有关指标出现明显变化,肾近曲和远曲小管出现超微结构病变,24h后出现光镜下病变,尿钠、钾排泄增加,尿渗透压降低。Ⅲ组灌胃后12h肾皮质匀浆自由基有关指标出现异常变化,肾近曲和远曲小管出现超微结构病变,晚于Ⅱ组出现;48h后出现光镜下病变,尿钠、尿钾排泄增加,尿渗透压降低。说明65个2对毒蕈所致大鼠急性肾损伤有一定的保护作用。

Acute kidney injury was induced by gentamycin peritoneal injection at the dose of 100 mg/kg for 10 days. It was observed that BUN, Scr,Upr and KI decreased significantly CMCs group compared with model group. Histopathological changes showed that renal tubule epithelial cell lesions were lessened obviously by microscope observation, and ultrastructural changes showed that lysosome and mitochondrion structural alterations were lessened markedly by TEM and SEM observation in CMCs group. 用庆大霉素ip造成大鼠急性肾损伤模型,观察到人工虫草可使尿素氮(BUN)、血清肌酐(Scr)、尿蛋白(Upr)和肾指数(KI)较病模组显著性下降,光镜检查显示人工虫草可使肾小管上皮细胞的肿胀、坏死等组织病理改变明显减轻,透射电镜(TEM)和扫描电镜(SEM)下可见细胞中溶酶体的改变和线粒体的损伤等超微结构变化亦显著减轻,上述结果提示人工虫草可改善肾功能,减轻肾水肿,保护庆大霉素对肾小管上皮细胞的损伤。

The acute pathogenetic effects of free radicals were investigated on isolated low-perfused kidney (IPK) during reperfusion and the protective effect of Cachannel blocker m-Nisoldipine was examined. The activity of SOD in renal cortex was reduced significantly with I hour low pressure perfusion(2. 0 ~2. 7kpa) followed by higher one (13. 3kpa ), but the contents of MDA, Ca and perfusing resistance were increased significantly. m-Nisoldipine could prevent these disturbances in dose-dependent manner.

本文采用离体低灌再灌注肾技术探讨氧自由基在急性肾损伤中的致病作用以及钙拮抗剂间尼索地平(m-Nis)的保护作用。低压(2.0~2.7kPa)灌注1小时后,继以13.3kPa灌注1小时,肾皮质氧自由基清除剂超氧化物歧化酶(SOD)活性明显降低,自由基代谢产物两二醛(MDA)、Ca含量及肾灌注阻力明显增加;低灌后再灌注时投以不同剂量的间尼索地平(0.05μmol/L与0.1μmol/L)呈剂量依赖性地提高SOD活性,降低MDA、Ca含量及肾灌注阻力。
