By means of high efficient method of cDNA synthesis directed by mRNA template, We have got a cDNA clone pFAS208 containing cDNA insert 3.7kb long for the gene fragment of rat liver fatty acid synthetase (RLFAS). pFAS203 clone has PstI, BamHI, HincII, PvuII, Aval and Pvul restriction enzyme sites. It is partial overlap with pFAS15 identified by hybrid-select and hybrid-arrest translation according the restriction enzyme mapping and Southern analysis of FAS cDNAs.The injection of insulin into fasted diabetic rat and refeeding with fat-free diet increased the amount of FAS mRNA and the nuclear precursor in the rat liver. The amounts of FAS mRNA and its precursor derived from diabetic rats induced with insulin, and refed fat-free diet for 12h is almost 30-fold of that from diabetic rats. Poly(A) RNA Northern analysis showed that the amount of FAS mRNA increased during insulin induction but the size of FAS mRNA kept in unchanged .This result demonstrate that insulin has effect on the regulation of transcription for FAS gene. The change of FAS activity during insulin induction is at the transcriptional level.

The inactivation kinetics of fatty acid synthctase(FAS) by modification of six SH reagents has been studied. It is found that the type of kinetics, the specificity of modification and protection by Salt or NADPH, are decided only by the carboxyl anion in reagent. It has been suggested that a carboxyl anion loading site exist in condensation center of FAS. This site is very likely the loading site of malonyl which contains a carboxyl anion, and is essential to condensation.

本文对六个修饰巯基的试剂失活脂肪酸合成酶(FAS)的动力学进行了系统研究,发现它们与FAS的作用是否属于亲和标记,修饰巯基时的专一性,以及盐和NADPH有无保护,均只与该试剂分子中有无羧基负离子有关,提出在FAS的缩合中心存在一个对羧负离子有特异亲和力的结合部位.该部位可能是丙二酰基在缩合中心的结合部位,为缩合反应所必需,它使携带了丙二酰基的活臂向缩合中心运动,并结合于此进行缩合反应.
