To detect viral genome in neurons of ganglia, tow step blot hybridization was applied in which all the necessary virual genome framgments were cloned into a single vector. The high titers of infectious HCMV could be detected by decreasing the incubation temperature from 40.5℃ at any time during the latent period. To determine the number of latently infected cells maintained at 40.5℃, infectious center assays were carried out. The infectious center titers ranged from 0.0004%—0.005%. The results showed a low reactivation rate.
