The early observation that adenylylation/deadenylylation was a major aspect of regulation of glutamine synthetase activity in Rhodopseudomonas capsulata was confirmed. When cetyltrimethylammonium bromide was used to stabilize the adenylylation state, a striking difference was observed between cells grown in ammonium, in glutamate and in N_2 with respect to the apparent adenylylation state of glutamine. The early observation that adenylylation/deadenylylation was a major aspect of regulation of glutamine synthetase activity in Rhodopseudomonas capsulata was confirmed. When cetyltrimethylammonium bromide was used to stabilize the adenylylation state, a striking difference was observed between cells grown in ammonium, in glutamate and in N_2 with respect to the apparent adenylylation state of glutamine. Glutamine synthetase was largely adenylylated in cells grown with an excess of ammonium, and deadenylylated when glutamate or N_2 was used as nitrogen source. Furthermore, glutamine synthetase activity in vivo was markedly stimulated by illumination. This effect was thought to be mediated, at least in part, by the change in level of ATP caused by photophosphorylation. The light-dependent nitrogen fixation as measured by acetylene reduction in Rps. capsulata was completely inhibited by the addition of a small amount of ammonia or by sudden interruption of illumination. The inhibition caused by ammonia was abolished by methionine sulfoximine, a glutamine synthetase inhibitor. The inhibition caused by darkness was, however, unaffected, indicating that different mechanisms were involved in the two short-term reversible regulation of nitrogenase activity. It seems likely that glutamine synthetase was involved in the short term regulation of nitro-genase activity caused by ammonia.

研究了荚膜红假单孢光合细菌菌株N-3谷氨酰胺合成酶(GS)的腺苷态/脱腺苷态间互相转换和调节方式,对十六烷基三甲基溴化铵添加后的稳定,以及不同氮源对酶量的变化和腺苷态的影响。在光暗的调节上,光强在500~50001x范围内,此酶的活性几乎处于同一水平,其腺苷态和脱腺苷态之比亦不发生显著变化,但一经黑暗处理,GS活性迅速下降,相似于固氮酶活性受光、暗的调节。借助于谷氨酰胺合成酶的抑制剂MSX加入反应系统,消除了氨对固氮酶活性的瞬间抑制,但消除不了暗对固氮酶活性的瞬间抑制,表明谷氨酰胺合成酶参与了氨对固氮酶活性的瞬间调节,但并不参与固氮酶活性的光,暗瞬间调节。
