The induction of hydrogenase preceded in the exponential phase of cultures, and all usable carbon substrates tested were found to repress the induction of hydrogenase. An effect of the addition of KNO_3 or NH_4Cl on hydrogenase formation was observed....

The expression of a H_2-uptake system in free—living cultures of Rhizobium astragalus occurred under heterotrophic growth condition in liquid medium. The expression was influenced by environmental factors, such as carbonhydrate, oxygen and hydrogen concentrations. High rates of H_2 uptake by R. astragalus were found by incubating bacterial cells in liquid or solid Hup medium under a gas phase of 40% N_2, 14%H_2, 4%O_2 and 4.2%CO_2. The cultures incubated under these conditions with the exception that Ar replaced H_2 showed no hydrogenase activity. When H_2 was removed after the onset of enzyme derepression, the hydrogenase activity did not increase further. High oxygen concentration repressed hydrogenase formation. The addition of chloramphenicol to derepressed cultures resulted in inhibition of enzyme formation similar to that of the repression by O_2

One Hup~+ strain was isolated from 68 free living strains of Rhizobium distributedover the south of Zhunkeer Basin in Xinjiang. Thestrains devoid of hydrogenase expression in free living state, showed hydrogenaseactivities after symbiosis with host plant.
