Therefore, according to the experimental curve of conversion in the reservoir, the mechanism of enzyme reaction becomes clear and the relevant parameters can be evaluated. With this method, the product glucose inhibition mechanism of maltose hydrolysis by glucoamylase, bound covalently to apolymethyl acrylate matrix, has been proposed, and its inhibition constant K_I, the apparent Michaelis-Menten constant K_M and the specific activity■ have been found.

用蟹壳、明胶和卡拉胶做载体,采用交联法和包埋法对糖化酶As3.4309进行固定化。以淀粉溶液做底物,在相同条件下,对各种固定化糖化酶进行活力测定。实验结果表明:用卡拉胶包埋的固定化糖化酶保留了最高、的酶活力;而酶活力半衰期最长的是用蟹壳与戌二醛交联的方法制得的固定化糖化酶,对所获得的结果进行了分析,从而为固定化酶的进一步研究提供了方法上的依据。
