Objective To study the effect of electromagnetic irradiation on steroidogenic acute regulatory protein (StAR) and cytochrome P450 cholesterol side chain clevage enzyme (P450 scc) in adult rat testis tissue.Methods The contents of testosterone in serum of rats at 3,6,24,72 h after electromagnetic irradiation and control rats were measured by using radioimmunoassay(RIA), meanwhile the levels of StAR mRNA and P450scc mRNA in testis tissue were measured by using semi-quantitative RT-PCR. Results The levels of testosterone, StAR mRNA and P450scc mRNA at 3 h after electromagnetic irradiation decreased 83.1%,57.3%,53.6% more respectively than those in control rats,(P<0.01). At 6 h, although those had a little elevation, still decreased 52.6%,17.9%,29.2% more respectively than in control rats ,(P<0.01,0.05), and recovered at 24 h. At 72 h, the levels of testosterone, StAR mRNA and P450scc mRNA fell again (decreased by 57.6%,39.5%,53.5% respectivly,P<0.01). Conclusion Electromagnetic irradiation may affect the transcription of StAR and P450scc in rat Leydig cells,thereby decrease testosterone synthesis.

目的探讨电磁辐射对成年大鼠睾丸组织中类固醇合成急性调节蛋白 (StAR)、细胞色素P4 5 0胆固醇侧链裂解酶 (P4 5 0scc)的作用。方法采用放射免疫方法分别测定电磁辐照后 3、6、2 4、72h及对照组血清睾酮含量 ,同时运用RT -PCR方法测定睾丸组织中StARmRNA、P4 5 0sccmRNA水平。结果辐照后 3h血清睾酮含量和StARmRNA、P4 5 0sccmRNA水平分别较对照组降低 83.1%和 5 7.3%、5 3.6 % ,(P <0 .0 1) ;6h略有回升 ,但仍分别较对照组降低 5 2 .6 %、17.9%、2 9.2 % (P <0 .0 1,P <0 .0 5 ) ;2 4h恢复到正常水平 ,72h再次分别较对照组降低 5 7.6 %、39.5 %、5 3.5 % (P <0 .0 1)。结论电磁辐射能在转录水平影响睾丸间质细胞StARmNA、P4 5 0sccmRNA的表达 ,从而降低睾酮的合成

OBJECTIVE:To study the effect of electromagnetic irradiation on steroidogenic acute regulatory protein(StAR) and cytochrome P450 cholesterol side-chain cleavage(P450scc) in the tissues of the testis of the adult rats,and to preliminarily reveal the molecular mechanism of electromagnetic radiation that affects the synthesis of testosterone. DESIGN:A randomized controlled study. SETTING:Department of Nuclear Medicine and Staff Room of Labour Hygiene, Institute of Field Surgery,Daping Hospital of the Third Military Medical University of Chinese PLA. PARTICIPANTS:The study was completed in the Department of Nuclear Medicine and Staff Room of Labour Hygiene, Institute of Field Surgery,Daping Hospital of the Third Military Medical University of Chinese PLA from June 2003 to May 2004.A total of 75 male Wistar rats of clean degree II,weighing 180 to 220 g,were randomly divided into 5 groups:control group,and groups of 3,6,24,72 hours after electromagnetic irradiation with 15 rats in each group. INTERVENTIONS:The rats were exposed to microwave with the peak power of 90 W/cm2 for 15 minutes successively,and all the rats survived the irradiation.The serum contents of testosterone of rats in the groups of 3,6,24,72 hours after electromagnetic irradiation and control group were measured by using radioimmunoassay(RIA) respectively;meanwhile the levels of the StAR and P450scc mRNA in testis tissue were also determined with RT-PCR. Objective To study mRNA expression of steroidogenic acute regulatory protein(StAR) and cytochrme P450 cholestero side chain lyase(P450scc ) in testicle and excretion of testrone in blood of rats post microwave exposure ,and to interpret the molecular mechanism of microwave exposure on the sexual fu nction in adult rats.Methods Capture latent period(CLP)and capture times(CT) w ere used to estimate sexual function of exposed rats.Peverse transcription-poly merase chain(PCR)reaction method was used to determine the expression of StAR,P 450scc mRNA in testicle of rats 3,6,24,72h post microwave exposure and meanwhile testrone lever in blood serum was measured by radioimmunoassay.Results Prolonged CLP and decreased DT were found after 3 -hour exposure(P<0.01 respectively) and sexual behavior came down signific antly.Testrone and StAR,P450scc mRNA changed in the same situation,compaed with control and dareased 83.1%,57.3%,53.6% respectively(P<0.01) after 3-h our exposure and recovered to normal in 24h but reduced once again after 72h and were decreased 57.6%,39.5%,53.5% respectively(P<0.01).Conclusion Leydig cells can affect and decrease the ex pression of StAR,P450scc mRNA and testosterone synthesis,and further to influenc e the sexual function of male rats exposed to microwave.
