This paper describes a series of studies on the comparison of the es-terase and peroxidase isozymes at the different developmental stages fromstamen,pistil and flag leaf of fertile and sterile plants of Tai-gu malesterile wheat,and of Timopheevi cytoplasmic male sterile line as well asa wheat ctltivar Nongda 139(as a control).The regulatory changes oftwo kinds of isozymes in different organs have been found.These twoisozymes in the stamen between the ferfile and sterile plants of Tai-gumale sterile wheat had the distinct differencs starting from the meiosis ofPMCS.The results showed that the time of appearance of the the unusu-al isozymes in stamen of sterile plants was in accordance with that of themicrogametic abortion.It was also found that the two isozymes in pistilwere different in early stage but identical in later stage,whereas,theisozymes in flag leaf had no difference to be seen.The appearance of above mentioned unusual isozyme indirectly indi-cated the existance of Tal gene in the sterile plants of Tai-gu wheatfrom the molecular level phenotype of gene expression.Whether unusualisozyme in the different organs existed is of certain significance to studythe Tal gene effects on the expression of the esterase gene and the pe-roxidase gene.

本文通过对太谷显性核不育小麦的可育株、不育株及 T 型不育系小麦和农大139小麦(可育株对照)的雄蕊、雌蕊及旗叶、不同发育时期的酯酶和过氧化物酶同工酶进行了一系列比较研究,找出了两种同工酶在各器官组织中的变化规律,发现了太谷可育株与不育株之间、雄蕊的两种同工酶从“花粉母细胞减数分裂期”时起均已有明显差异。说明不育株雄蕊中异常同工酶产生的时期与其细胞形态学观察到的小孢子败育时期一致。还发现雌蕊的两种同工酶在前期也有差异,但后期趋于一致。旗叶的同工酶尚未见差异。上述异常同工酶的出现是从基因表达的分子水平表型,间接证明了太谷不育株中 Tal 基因的存在。异常同工酶在不同器官组织中的有无,对于研究 Tal基因对酯酶基因表达和对过氧化物酶基因表达的影响具有一定意义。此外,本文还对 T 型不育系小麦与太谷核不育小麦的酯酶及过氧化物酶同工酶,进行了初步比较研究。

Three organophosphate resistant strains(FS-1, FS-2 and CD-1) were selected from collections of Culex quinquefasciatus from Foshan and Chengdu, China. Different esterase patterns were observed in the three strains. The numbers of esterase B gene copies were approximately 32-fold in FS-1, 8~16-fold in FS-2 and 2~4-fold in CD-1. The amplification levels of esterase B genes were correlated with esterase activities and OP resistances among the three strains.The restriction pattern comparison of the amplified region containing the esterase B gene revealed large differences among the three strains. The esterase B1 was found in FS-1 and two esterases, B2 and a new B haplotype (named B6), were detected in FS-2. Another new haplotype(named B7)was observed in CD-1. Esterase B6 and B7 had identical electrophoretic mobility, but had different restriction patterns within the regions flanking the esterase B gene, indicating that two distinct haplotypes were amplified independently in Culex populations in Foshan and Chengdu,China.

in Culex pipiens, overproduction of nonspecific esterases is a commonmechanism of resistance to organol,hosphate insecticides. The esterases are attributed to closely linked loci named A and B, and overproduction of all esterases B is due to gene amplification. In order to determine whether the esterase Bl identified by electrophoretic studies in Culex pip lens mosquitoes from different countries is over-produced due to the amplification of the same DNA haplotype, the amplified region encompassing the structural esterase Bl gene was characterized by restriction mapping and RFLP. Thesame amplified haplotype was found in mosquitoes with an esterase Bl protein,independently of their geographical origin: French Guyana, Venezuela, Puerto Rico,California and Chuna. Large variations in amplification levels were observed. In addition to the highLly amplified 2.Ikb fragment, EcoRI digestion led to two other restriction fragments of low amplification in mosqintoes from the Americas but not from China. It is concluded that BI amplification has a unique origin, either in America or in Asia, and has subsequently spread on a large scale by migration. Thismigration is more limited than that of A2-B2 esterases, since Bl is confined to the Americas, Carribean and part of China, whereas the A2-B2 distribution now includes the Americas, Carribean, Asia, Africa, Pacific islands and Europe.

在库蚊Culexpipiens品系中,非专一性酯酶的过量产牛是对有机磷杀虫药剂抗性的普遍机理。酯酶基因位于紧密连锁的A和B座位上。现已知所有酯酶B的过量产牛都是基因扩增的结果。为了确定不同国家库蚊品系的酯酶B1的过量产生是否都是相同DNA单基因型扩增的结果,我们构建了酯酶B1结构基因扩增区的限制性内切酶酶切图谱,分析了限制性酶切片段长度多态性(RFLP)。研究发现不同地理位置的酯酶B1库蚊,如法属圭亚那、委内瑞拉、波多黎各岛、美国加利福尼亚和中国北京,都有着相同的单基因扩增,但在扩增水平上有较大的差异。我们认为无论在美洲或亚洲,凡是酯酶B1扩增的库蚊都为同一个起源,之后经迁移而传播到各地;同时发现酯酶B1扩增的库蚊与酯酶A2-B2扩增的库蚊相比,其迁移有一定的局限性;并巨酯酶B1扩增的库蚊仅仅限于美国、加勒比和中国的一些地区,而酯酶A2-B2扩增的库蚊则广泛地分布于美国、加勒比、亚洲、非洲、太平洋各岛及欧洲等地。
