These structures are the presynaptic axon and various parts of the postsynaptic neuron. Aside from the usual forms of the response of these structures, less usual forms were described. Single and repetitive stimulations of the optic nerve by electrical shocks were employed as a means of differentiating pre- and post-synaptic response. (2) In substantiating a view expressed by some workers, further evidence has been adduced to indicate that activity due to a single optic nerve fibre is capable of initiating postsynaptic discharge. However, the readiness of different neurones to fire upon the arrival of impulses from the optic tract is quite variable. In general, this variability is due not so much to the smallness of excitatory synaptic potential initiated as to the condition imposed by the mixed action of both excitatory and inhibitory input. (3) Two steps in the generation of the postsynaptic spike could be dissociated by experiment using two antidromic shocks. This offers a direct proof that the two steps are respectively generated by the initial segment of the axon and the neuron soma proper.

This slow potential was resistant to d-tubocurarine (d-TC) and atropine,and could be reversibly blocked by a low calcium (0.25 mM) and high magnesium12 mM) Krebs solution; consequently the potential was termed non-cholinergicexcitatory postsynaptic potential and could be categorized as a kind of late slow ex-citatory postsynaptic potential (ls-EPSP). The ls-EPSP in the majority of cellssampled was accompanied by an inrease in membrane resistance and was attenu-ated upon conditioning hyperpolarization, which suggested that an inactivationof potassium conductance was probably involved in genesis of the slow potential.Superfusion of substance P caused no appreciable change in the ls-EPSP. In addi-tion, some of the cells in the rabbit IMG generated a hyperpolarization potentialafter a train stimulation of the preganglionic fibers. The hyperpolarization lastedabout 1 min and was enhanced by conditioning hyperpolarization. It was alsod-TC and atropine resistant, but sensitive to low calcium/high magnesium; there-fore, it was termed "very slow inhibitory postsynaptic potential" (vs-IPSP) to bedistinguished from the s-IPSP, a conventional term for the slow cholinergic synapticresponse in some autonomic neurones. The transmitters responsible for these twotypes of non-cholinergic synaptic potentials remain to be identified.

In addition to antidromic, orthodromic, and spontaneaus potentials regularly recorded,synaPtid potentials were also registered.No distinct prepotentials and A-B in flection were seen in the antidromic potentials. Each orthodromic, spontaneous, and synaPtic potential consisted of a prepotential, a spike, and a afterpotential and showed A and B components on the rising phase of their spike. Postsynaptic potentials manifested current effect and frequency dependency and their IPSP displayed a prolonged inhibition.The results indicate that dorsal column postsynaptic neurons possess a relatively simplified geometric organization and a considerable complexity in their local synaptic circuitry.
