Objective To investigate the cooperation effect of p33 ING1b , coded protein of the novel tumor-suppressor gene ING1b, with p53 gene on the cell growth, cell cycle arrest, apoptosis and expression of the p53 downstream gene p21 WAF1/CIP1 in hepatocellular carcinoma.
The authors analyse DNA binding sequence for wild-type P53 in human genome by Computational method and discover that there is a consensus sequence,5'-GGGCTAGTTTtgGAGCTAGCCT-3',in intron 2 of the TSPY1(testis specific protein,Y-linked 1) genes in hunan chromosome Y. It suggests that TSPY1 gene may be a p53 downstream gene.
Results Genistein could inhibit proliferation of human breast carcinoma cells with different ER status and p53 status. This inhibition was associated with induction of p21 WAF1/CIP1 mRNA and protein expression. Exposure of MCF 7 and MDA MB 231 cells to genistein resulted in statistically significant increased p21 WAF1/CIP1 mRNA stability, accompanied with increased p21 WAF1/CIP1 protein expression.
