Objective: To study the role of the apolipoprotein J (apoJ) gene in the genetic background of pregnancy induced hypertension (PIH). Methods: The exons of apoJ gene were screened for the presence of the polymorphic variants in 50 cases of pregnancy induced hypertension and 50 cases of normal pregnant women with PCR DGGE technique. Objective: To study the role of the apolipoprotein J (apoJ) gene in the genetic background of pregnancy induced hypertension (PIH). Methods: The exons of apoJ gene were screened for the presence of the polymorphic variants in 50 cases of pregnancy induced hypertension and 50 cases of normal pregnant women with PCR DGGE technique. Results: A total of 38 cases of heterozygote and 1 case of homozygote about the exon5 of apoJ gene were observed. Among them, 25 heterozygotes and 1 homozygote were found in the control group. 13 heterozygotes were in the PIH group. There was one homozygote of apoJ gene exon7. The heterozygous variant frequency of apoJ gene exon5 in PIH group (26%) was significantly lower than that in normal pregnancy group (52%) ( P<0 01 ). No other abnormal bands were observed for others exons of apoJ gene. Conclusion: The polymorphic variation of exon5 of apoJ gene might associate with PIH.[

目的 :探讨载脂蛋白J(apoJ)基因在妊娠高血压综合征 (妊高征 )遗传背景中的地位。方法 :运用聚合酶链反应 变性梯度凝胶电泳 (PCR DGGE)技术对 5 0例妊高征患者和 5 0例正常孕妇的apoJ基因外显子进行了多态变异筛查。结果 :对apoJ基因 5号外显子筛查 ,共检出 38例杂合子和 1例纯合子 ,其中正常孕妇组有 2 5例杂合子和 1例纯合子 ;妊高征组有 1 3例杂合子 ,有 1例的 7号外显子也为杂合子。 5号外显子的杂合子多态变异基因型频率在妊高征组为 2 6 % ,显著低于对照组的 5 2 % (P <0 0 1 )。其余外显子DGGE未检出异常电泳条带。结论 :apoJ基因 5号外显子变异可能与妊高征的发病有关

Objective To investigate the mechanism of the neuro protective effect of apoJ in rat acute cerbral infarction.Methods 84 healthy male SD rats were devided randomly into normal control group,sham operated group,cerebral infarction 6 hours group,24 hour group,3 day group,5 day group and 7 day group.Permanent cerebral infarction models were prepared by occluding the middle cerebral artery (MCA) using thread embolizing.Brain water content (BWC) was measured,and the C3 and apoJ mRNA expression was analyzed by RT PCR.Results A low level expression apoJ mRNA was detected in normal rat brain and an increased expression was found in ischemia rat brain.A negative correlation was found between BWC and apoJ mRNA expression and a positive correlation between C3 mRNA and apoJ mRNA expression ( r = -0.212 and 0.509 respectively, P < 0.05 and P < 0.01 ).Conclusions Increased apoJ expression may be invoved in neuroprotection in cerebral infarction by inhibiting the complement activation.

目的 探讨载脂蛋白J (apoJ)在大鼠脑梗死急性期的作用及其可能的机制。 方法 清洁级SD雄性成年大鼠 84只 ,随机分为正常对照组、假手术组、脑梗死后 6h组、2 4h组、3d组、5d组和 7d组。用线栓法制备永久性局灶脑梗死模型 ,动物处死后测脑含水量 ,并检测apoJ及补体C3mRNA的表达值。 结果 对照组大鼠脑内有apoJmRNA表达 ;各模型组缺血侧ApoJmRNA表达增高。ApoJmRNA表达值与脑含水量呈负相关 (r=- 0 .2 12 ,P <0 .0 5 ) ,与补体C3mRNA表达水平呈正相关 (r =0 .5 0 9,P <0 .0 1)。结论 大鼠脑梗死后apoJ表达增高 ,3d达高峰。ApoJ可能有保护缺血脑组织的作用。
