Purpose To study the expression of human papilloma viruses 16 and 18 (HPV16/18) DNA and the cell cycle proteins of p16~ Ink4a and Rb in cervical adenocarcinoma and the influence of HPV16/18 infection on expressions of p16~ Ink4a and Rb protein.
Methods A549 cell line was transfected with p16 INK4A and the stably transfected strain was identified with Northern blot and Southern blot. Cell cycle was analysied by flow cytometry. The phosphorylation state of retinoblastoma protein(Rb) and cell phenotype were determined by Western blot and SA-β-gal staining.
Rb gene protein positive expression rate in tumor tissues was 51.4%,which was also lower than that in normal mucosal tissues of colon and rectum (χ2=4.234,P< 0.05); Rb protein positive expression correlated significantly with lymph node metastasis(χ2=14.596, P< 0.05), but did not correlated with tumor cell differentiation( P >0.05).
