Purpose] To investigate the inhibitive effect and related factors of the γ-interferon(IFN-γ) transgenic vaccine on xenogeneic S180-bearing mice. [Methods] IFN-γ gene transduced vaccine was obtained from IFN-γ transformation cell line after 60Co radiation. MHC-Ⅰ?Ⅱ molecule expressing levels were detected and their changes were evaluated by means of the radiation and freezing. Inhibition effect on the xenogeneic S180-bearing mice was evaluated after changing the injection site and adding GM-CSF, IL-2 and tumor cell lysates. [Results] The cell line could express high MHC-Ⅰ?Ⅱ levels after being transduced by IFN-γ gene which were not affected under 60Co radiation and freezing condition. The inhibition affects were not effected after changing the injection site and using IL-2 as a replacing transgenic vaccine. [Conclusion] The results suggest that the IFN-γ transgenic A549 cell vaccine under different condition such as: radiation and freezing, or change injecting site, or using IL-2 to preplace the transgenic vaccine could still have a strong inhibitive effect on the xenogeneic mouse tumor.

[目的]探讨γ鄄干扰素(IFN鄄γ)转基因疫苗对异种小鼠S180肿瘤的抑瘤作用及其影响因素。[方法]将IFN鄄γ基因修饰的转化细胞株,经γ鄄60Co照射后制成转基因疫苗,测定其MHC鄄Ⅰ、Ⅱ分子的表达,以及60Co照射与冻融等因素对其影响。在添加肿瘤细胞裂解物和GM鄄CSF、IL鄄2及变换注射部位等,观察对异种荷瘤S180小鼠的抑瘤作用。[结果]经IFN鄄γ基因修饰后,其细胞表达MHC鄄Ⅰ、Ⅱ分子明显提高,该两分子的上调并不受照射及冻融等因素的影响。变换注射部位及用IL鄄2取代转基因疫苗作预防性注射后依然有抑瘤作用。[结论]经IFN鄄γ基因修饰的A549细胞疫苗其MHC鄄Ⅰ、Ⅱ分子的上调并不受照射、冻融等因素及变换注射部位的影响,仍可对异种动物肿瘤产生较强的免疫抑瘤作用。

猪传染性胃肠炎病毒纤突蛋白是诱导猪产生特异性中和抗体的主要结构蛋白,包括A,B,C,D4个抗原决定位。为进一步生产TGEV植物转基因疫苗提供可操作性强、具免疫活性的外源基因,以猪传染性胃肠炎华毒弱株(TGEVH)的S基因为基础,扩增并连接4个抗原决定位相应的碱基序列,构建pPROEX-HTa原核表达载体,转化大肠杆菌诱导表达。结果表明,修饰后的S蛋白的表达量为19.8%,仍能和相应的血清发生抗原抗体反应。
