[Objective]To study the using of fragile histidine triad (FHIT) gene in molecular staging of non-small cell lung cancer (NSCLC).[Methods]64 stations of lymph nodes from 28 patients with NSCLC were studied to detect mRNA for FHIT gene by using reverse transcription-polymerase chain reaction assay (RT-PCR).[Results]Of the samples of lung and the lymph nodes of the patients without lungcancer,the FHIT gene was expressed at mRNA level in 0%.The expression rate in samples of lymph nodes using RT-PCR (37.5%,24/64)...

[Objective]To study the using of fragile histidine triad (FHIT) gene in molecular staging of non-small cell lung cancer (NSCLC).[Methods]64 stations of lymph nodes from 28 patients with NSCLC were studied to detect mRNA for FHIT gene by using reverse transcription-polymerase chain reaction assay (RT-PCR).[Results]Of the samples of lung and the lymph nodes of the patients without lungcancer,the FHIT gene was expressed at mRNA level in 0%.The expression rate in samples of lymph nodes using RT-PCR (37.5%,24/64) was significantly higher than that using routine histopathological examination (26.6%,17/64).[Conclusion]There were loss and transcript abnormalities of FHIT gene in NSCLC.FHIT loss and transcript abnormality preferentially occur in squamous cell carcinomas. The establishment of FHIT may help for molecular staging of lung cancer.

[目的]研究脆性组氨酸三联体(fragilehistidinetriad,FHIT)基因在非小细胞肺癌(NSCLC)分子分期中的应用。[方法]采用逆转录-聚合酶链反应(RT-PCR)方法检测28例患者的64组淋巴结FHIT基因mRNA的表达。[结果]非肺癌患者淋巴结FHIT基因失表达或异常表达(阳性表达)率为0,应用常规病理检查和应用RT-PCR技术检测FHIT基因在淋巴结微转移阳性率分别为26.6%(17/64)和37.5%(24/64),差异有显著性(P<0.05)。[结论]非小细胞肺癌淋巴结中存在FHIT基因失表达或转录表达异常,检测肺癌淋巴结中FHlT基因的表达对准确判断肺癌的分子分期有帮助。

The molecular technique was used in the molecular staging of lung cancer to find the occult micrometastasis of the tumor, and to forecast the metastasis and recrudescence of the tumor. The method was used for checking genetic mutations or protein expressions in the lymph node, blood and bone marrow of patients with cancer. The article dissertates the concept of molecular techniques and the result in molecular staging of NCLC. It indicates the predominance and deficiency of the molecular techniques in present study.

癌症的分子分期(molecularstaging)是指应用各种先进的分子生物学诊断技术检查癌症患者的淋巴结、循环血液及骨髓等组织,从中发现常规影像学或病理组织学无法检测到的隐性微小转移灶,进而从基因或蛋白质水平诊断癌症的转移,根据微转移发生的部位,结合癌症的国际分期标准,最终达到更准确的TNM分期。本文介绍了癌症分子分期的概念,对国内外学者在非小细胞肺癌的分子分期的研究工作进行综述,并提出目前研究中运用的分子生物技术的优势与不足。
