In the present study the influence of melatonin on the expression c-fos in primarily cul-tured kidney epithelial cells of duck was studied by immunocytochemistry.Kidney cells from sev-en week old duckswere isolated by a mechanical mcthod and the cells were cultured inDulbecco's Modified Eagle's Medium(DMEM) with 10% fetal calf serum for 48 hours,then infetal calf serum free DMEM for another 24 hours;In the experiment the cells were divided intocontrol group,10% fetal calf serum stimulated group and melatonin 10-6mol/L plus serumgroup,The results of the experiment showed that Fos immunoreactive cells increased to 63.07 ±3.93% after exposure to fetal calf serum,significantly higher than that of the control group(44.31±2.77%,P<0.01).In the melatonin plus serum group the Fos immunoreactive cells decreased to35.29 ± 3.22%,which was significantly lower than that of the serum treated group(P<0.01),butnot statisticalIY different from that of the control group.The results suggest that melatonin inhib-its the expression of c-fos in cultured kidney epithelial cells of duck induced by fetal calf serumstimulation.

本研究采用Fos蛋白免疫组化染色方法观察了胎牛血清对原代培养鸭肾上皮细胞c─fos表达的刺激作用及褪黑素的阻断作用。以每一视野Fos染色阳性细胞的百分比为指标,每组实验观察14至17个视野。实验结果显示,正常对照组平均每视野Fos染色阳性细胞为44.31±2.77%,胎牛血清刺激组为63.07±3.93%,明显高于对照组(P<0.01),胎牛血清加10 ̄(-6)mol/L褪黑素组为35.29±3.22%,明显低于单纯胎牛血清刺激组(P<0.01),但与对照组无明显差异(P>0.05)。该结果表明褪黑素可以抑制原代培养鸭肾上皮细胞c─fos表达。

AIM To observe the protective effect of Qinling chongji (QL) on liver and renal functions, and the effect on blood biochemistry in DHBV induced hepatitis nestling ducks. METHODS When positive serum DHBV was detected in seven day infected nestling ducks, animals were randomly divided into five groups: ①normal saline (NS); ②positive control drug, ACV 100*!mg·kg -1 . ③ QL 10*!g·kg -1 (QLb); ④QL 5g·kg -1 (QLm);⑤ QL 2 5*!g·kg -1 (QLs). Five to six animals were used in each group....

AIM To observe the protective effect of Qinling chongji (QL) on liver and renal functions, and the effect on blood biochemistry in DHBV induced hepatitis nestling ducks. METHODS When positive serum DHBV was detected in seven day infected nestling ducks, animals were randomly divided into five groups: ①normal saline (NS); ②positive control drug, ACV 100*!mg·kg -1 . ③ QL 10*!g·kg -1 (QLb); ④QL 5g·kg -1 (QLm);⑤ QL 2 5*!g·kg -1 (QLs). Five to six animals were used in each group. Drugs were administered orally, twice daily for 10 days. The blood samples were taken from leg vein 10 days after treatment and sera were separated at once. RESULTS Blood biochemical indexes of nestling ducks showed significant abnormality 7 days after DHBV infection, of which liver function (ALT, AST, SB)and renal function (UN, CR) changed the most, secondly,LDH level increased significantly. The animals were treated with different doses of QL for 10 days, above normal biochemical indexes were lowered in QLb (10*!g·kg -1 ) group ( P <0 05 , P <0 01 ). Liver pathological examination demonstrated that the constructions of liver leaflet in DHBV control group were damaged completely, while those in QLb group maintained intact. CONCLUSION QL can protect liver tissues in DHBV induced hepatitis nestling duck and could decrease the serum levels of ALT, AST, SB, UN and CR, especially the later two. The liver and renal functions were improved after treatment with QL, so it is worthwhile to perform further clinical studies on this anti hepatitis drug.

目的 观察芹灵冲剂(Qinling chongji, QL) 对DHBV 肝炎雏鸭肝脏的保护作用及对肝炎雏鸭肾功能、血液生化改变的影响.方法 待DHBV 感染雏鸭7 d 血清检测为阳性后,随机分组进行药物治疗试验,每组5 ~6 只,QL 分为高(QLb) 、中(QLm) 、低(QLs)3 个剂量组,分别为10 ,5 和2-5 g/ kg 组,ig ,2 次/ d ,10 d . 设病毒对照组(DHBV) ,以生理盐水代替药物. 阳性药用阿昔洛韦( 无环鸟苷,ACV) 粉剂100 mg/ kg ,ig .2 次/ d,10 d ,自鸭腿胫静脉取血,分离血清,用全自动生化测定仪分别测定肝功能(ALT,AST,SB) 、肾功能( UN,CR) 及其他生化指标的含量. 并将各组动物采血后剖腹取出肝脏,在肝脏同一部位取样,100 m L/ L中性福尔马林液固定,常规石蜡包埋切片,HE 染色,光镜观察.结果 雏鸭被DHBV 感染7 d 后血液生化指标明显异常,其中肝功能(ALT,AST,SB) 和肾功能(UN,CR) 改变最明显,其次乳酸脱氢酶(LDH) 显著升高. 用药组经不同浓度的QL 分别治疗10 d ...

An auto-reflut-digester is selected to digest duck kidney by the means Of HNO_3+H_20_2 (2 +1) and six elements (copper,zinc, lead, iron, calcium, cadmium) were determined by flame atomic absorption spectrometry (FAAS). The experiment showed the method was rapid and accurate. RSD (n=5) is 1.2%-10. 1% and ratio of recovery is between 84.00% -94.25%.

选择采用 HNO_3+H_2O_2(2+1)消化法对鸭肾进行消化处理,用火焰原子吸收光谱法测定鸭肾中铜、锌、铅、铁、钙、镉等六种元素,实验表明该法有耗样少、精确度高、快速等优点,结果令人满意,该法具有一定的实用意义。
