The protective effect of pretreatment with bismuth nitrate on acute renal toxicity by cisplatin was investigated in rats. Male rats received bismuth nitrate 30 μmol·kg-1 ·?d-1 sc for 3 d and a single dose of cisplatin 5 mg ·kg-1 ip on d 4. The pretreatment with bismuth significantly protected the rats from the cisplatin-induced rise in blood urea nitrogen and the widespread tubular necrosis in the S3 segment of the proximal tubule situated in the outer stripeof the medulla, although a minor transient increase...

The protective effect of pretreatment with bismuth nitrate on acute renal toxicity by cisplatin was investigated in rats. Male rats received bismuth nitrate 30 μmol·kg-1 ·?d-1 sc for 3 d and a single dose of cisplatin 5 mg ·kg-1 ip on d 4. The pretreatment with bismuth significantly protected the rats from the cisplatin-induced rise in blood urea nitrogen and the widespread tubular necrosis in the S3 segment of the proximal tubule situated in the outer stripeof the medulla, although a minor transient increase of protein and NAG in the urinary excretion was observed. In comparison with the control, a single dose of cisplatin was unable to induce the synthesis of renal MT, while the pretreatment with bismuth resulted in a 5-fold increase in the content of renal MT. The results suggest that MT preinduced by bismuth may play an important role in the protection against the acute renal toxicity of cisplatin.

本文在以往研究证实天然虫草对氨基糖甙肾毒性损害具有良好防治作用的基础上,以人工培植虫草替代天然虫草,观察它对体外培养肾小管上皮细胞增殖,对庆大霉素所致大鼠急性肾毒性损害及庆大霉素血药浓度的影响。结果表明,人工虫草可减轻庆大霉索对肾的损害,使大鼠尿 NAG 酶排泄量减少,保钠功能相对保存,血尿素氮、肌酐上升幅度小,其菊粉清除率高于单用庆大霉素对照组.人工虫草与庆大霉素联合应用既不影响庆大霉素血药浓度,也不减少肾皮质的庆大霉素积聚量,提示其保护肾脏的机理可能不是通过抑制庆大霉素转运和拮抗庆大霉素的药理作用。而主要是通过促进对损害、坏死的肾小管细胞再生和修复。

This study was carried out to observe the alteration of EGF in renal tissue and the amount of urinary EGF excretion in gentamicin toxicity rats with or without the supplementation of Cordyceps Sineasis (CS).Male SD rats were randomely divided into two groups.Each group recieved gentamicin 160mg/kg·Bw/day,ip×7 day.In adition gentamicn+CS group was adiministcred CS 1g/day p.o,×7 day and gentamicin group was given normel salin simultaneously.It was found that there were a redistribution of EGF in renal tubule and decrease of urinary EGF excretion during the acute phase of gentamicin toxicity.However the amount of identified in renal tubular cells and the excretion of EGF in urine increased progressively toward the recovery period of ARF.Interestingly,the changes of urinary EGF excretion,renal tubular regeneration and renal function recovery in gentamicin+CS group were more earlier and faster than that in gentamicin group.Our study suggested that CS might be by way of increasing urinary and renal tissue EGF to accelerate tubular repair and promote recovery of renal function.

本研究动态观察了冬虫夏草对肾毒性损害大鼠尿及肾组织中 EGF 变化的影响。实验结果表明,在急性肾毒性损害早期,肾组织中 EGF 出现重新分布,尿 EGF 排泄减少,而随着損害的修复,尿及肾组织中 EGF 逐渐增加;冬虫夏草治疗组大鼠尿及肾组织中 EGF 升高较庆大霉素组明显,且高峰提前,与此平行,肾小管修复较早,肾功能恢复较快。提示冬虫夏草增加 EGF 的作用,可能是它加速小管修复,促进肾功能恢复的机理之一。
