By the method of ultramicro-electrophoresis on cellophane paper,the contentsof free amino acids(FAA)in the motor fibers,the medial giant fibers andin the giant nerve cells isolated from ganglions of abdominal nerve cord in theshrimp were determined.The results obtained were as follows:1.The FAA contents in the motor giant fiber were much higher than thosein the medial fiber.The concentrations of aspartic acid,glutamic acid and totalneutral amino acids in the motor giant fiber were respectively 7.5,3.2 and 3.3times higher than those in the medial fiber.Moreover,in the motor giant fiberthe concentration of aspartic acid was twice as high as that of glutamic acid,whilein the medial giant fiber no such difference was found.2.The concentration of the acidic amino acids in these giant fibers of theshrimp was much lower than that in the nerve fibers of other marine animals,such as lobster,crab and squid.The concentration of the acidic,amino acidsin the medial giant fiber was even as low as that in the sciatic nerve of thetoad.3.The FAA contents in the abdominal ganglionic cells were much higherthan those in the giant fibers.A considerable amount of λ-aminobutyricacid(0.13%)was found in the cells,but no trace of it could be detected inthe fibers.

Thirty patients undergoing gastrectomy for benign lesions were divided into 3 groups.The 1st group received postoperatively a domestic product consisting of 18 amino acids(manufactured by Shanghai Institute of Pharmaceutical Industries),glucose and electro-lytes for 4 days.Their nitrogenous intake was 24 gm/day.They were compared with 10patients each receiving glucose-electrolytes and glucose-electrolytes-Proteamin 12X(a Tanabeproduct)respectively.The net nitrogen balance of the 3 groups during the 4 days were-32.08±5.03,-49.46±8.20 and -30.81±6.36,showing significant protein sparingeffect in the groups receiving amino acids.The serum free amino acids level and urinaryamino acids output showed no significant differences among the 3 groups.The dailyretention rates of domestic amino acids administered were 88.8%,86.4%,89.9% and88.4%.The protein sparing effect,urinary excretion and retention rate of the domesticproduct are comparable to that of proteamin 12X.However,serum bilirubin level rosefrom 0.55 to 1.07 mg%(P<0.05)with the use of domestic amino acids solution whileno such rise was observed with proteamin 12X.This tends to limit clinical applicationof this new product.

本文报道10名胃大部切除术患者应用“国产18种氨基酸注射液”的结果,术后4天的氮平衡、尿中游离氨基酸排泄量、血中游离氨基酸浓度变化的结果均与日本产品 Proteamin 12X无显著差异。血、尿常规,血尿素氮检查未发现明显改变。谷丙转氨酶及锌浊度试验均在正常范围内,输注后病人没有出现呕吐、恶心、皮疹或发热寒颤等不良反应。但有4例在输注后出现高胆红素血症(超过1毫克%),有待进一步改进。

So we have successfully tried to analyse free amino acids in plasma with reversed-phase high performance liquid chromatograhy, using gradient elution, o-phthalaldehyde / 2-mercaptoethanol...

Amino acids are usually determined by an ami-no acid analyser, the whole procedure consisting of separation of atnino acid mixtures by classical ion-exchange chromatography, post-column deri-vatization of ninhydrin and UV detection, is quite time consuming, especially in the determination of amino acids in physiological fluids. So we have successfully tried to analyse free amino acids in plasma with reversed-phase high performance liquid chromatograhy, using gradient elution, o-phthalaldehyde / 2-mercaptoethanol pre-column derivatization and fluorescence detection. The present method is able to determine twenty two plasma amino acids at one run within fifty minutes. The apparent advantages of this method are \ simple preparation, short period of derivatization, Small sample size, high precision and accuracy, and high recorvery of each amino acid(96.8%?.5%). We have examined experimentally the factors that could influence chromatographic separation, and disclosed suitable conditions for determination, such as program and volume of elution, property and temperature of the column, pH and ion concentration of buffer solutioa in the mobile phase, and the time and temperature of derivatization reaction.

本文报告了采用高效液相色谱法反相梯度洗脱,邻苯二甲醛和β-巯基乙醇柱前衍生化,萤光检测分析血浆游离氨基酸。实验采用线性洗脱,在50分钟内可同时测定22种氨基酸。血浆样品的预处理简单,衍生化反应的时间仅需1分30秒,血浆样品的实际进样量少于1微升.本测定方法的精确度高,各个氨基酸保留时间的变异系数平均为0.33%±0.26%(SD),峰面积的变异系数平均为1.72%±1.25%(SD),各个氨基酸的浓度在10~300毫微克/微升的范围中,线性关系的相关系数平均为0.996±0.007(SD).准确性亦好,各个氨基酸的回收率平均为96.8%±4.5%(SD).实验中还比较和讨论了梯度型式和洗脱体积,不同固定相和色谱柱温度,流动相中缓冲溶液的pH值和离子浓度以及衍生化反应的时间和温度等因素对分析结果的影响。
