Conclusion:The recombinant proteins were successfully expressed in BL21(DE3)and purified. This study provides necessary data for further investigation of the immunogenicity and anti-cancer vaccine of fusion protein hCGβ-(CTP)_4-(C3d-P28)_4.
Conclusion: The fusion protein hCGβ-(CTP)n(n=3,4)-3C3d was successfully expressed in Pichia pastoris. This study prepared basic necessary data for further investigation of the immunogenicity and anti-cancer vaccine of fusion protein hCGβ-(CTP)n (n=3,4)-3C3d.
Thesubclone DNA fragments with different function and better presentation of mimic antigen were inserted into thebacteriophage T7 expression vector. The anti-cancer vaccine of recombinant bacteriophages T7 were constructedand the mice were immunized. The specific antibodies and the cytotoxic T lymphocyte response were detected withdot blot and MTT method, respectively.
The results of ELISPOT and LDH release assay showed that MAGE3/HSP70 generated tumor specific cytotoxic T lymphocyte (CTL) to damage tumor cel1. CONCLUSION: High cell- densityfermentation of MAGE3/HSP70 engineered E. coli was realizedand MAGE3/HSP70 fusion protein can generate specific CTL to kill tumor cel1.
