In this paper, the amount of squalene in 7 deepwater sharks was discussed. In mo st of deepwater sharks, liver makes up around 20 percent of the total body weigh t. The liver of most of these sharks contains over 50 percent oil; some of them over 80 percent. The amount of squalene varied for individuals and species, but generally it was over 50 percent by weight. The preparation methods of squalene and squalane was also discussed.

Methods: Immunocompetence of shark liver extracts were measured respectively by 3H-TdR intermingle assay, cellular technique, ELISA, biotin-streptomycin labeled avidin (BSA) assay and 125I-UdR release test. Results: Shark liver extracts could significantly accelerate DNA synthysis of 7211 cells, enhance Ea- rossette forming cell rate of human peripheral blood mononuclear cells (PBMC) (P<0. 01), induce the secretion of IFN-γ by PBMc (P<0. 01), increase the activity of NK cells (P< 0. 05), and boost the expression of HLA-DR antigens (P<0. 001). Conclusion: Shark liver extracts can adjust positively the human cellular immunity. The results implied that it ha the higher values for application and research on approving immunologic conditions and enhancing homunologic defence.

鲨鱼肝脏中提取一组能有效促进成纤维细胞等多种组织细胞增殖、分化的小分子活性多肽,体外探讨其生物活性及其对人细胞免疫功能的影响。方法:分别用~3H-TdR DNA掺入法、细胞学技术、ELISA、生物素-链霉亲和素(BSA)免疫细胞化学法及~(125)I-UdR释放试验检测其免疫活性。结果:可明显增强肝瘤细胞株7211细胞DNA合成,有效提高T淋巴细胞形成Ea花环能力(P<0.01),诱导PBMC细胞分泌IFN-γ(P<0.01),增强NK细胞活性(P<0.05),促进PBMC细胞表面HLA-DR抗原的表达(P<0.001)。结论:鲨鱼肝提取液对机体细胞免疫功能具有良好的正向调节作用,提示其在改善机体免疫状态,提高机体免疫防御机制等方面具有较高的应用价值。

An active polypeptide ,named Shahepatide ,was extracted and purified from Shark liver. This article investigated its therapeutic effects on chronic liver injury induced by CCl 4 in rats. The results showed that Shahepatide could suppress the elevation of ALT, AST and hydroxyproline in serum of liver injury rats induced by CCl 4, and could increase albulmin level and the ratio of albulmin and globulin in serum. An active polypeptide ,named Shahepatide ,was extracted and purified from Shark liver. This article investigated its therapeutic effects on chronic liver injury induced by CCl 4 in rats. The results showed that Shahepatide could suppress the elevation of ALT, AST and hydroxyproline in serum of liver injury rats induced by CCl 4, and could increase albulmin level and the ratio of albulmin and globulin in serum. Hepatic histological findings indicated that Shahepatide could alleviate hepatic fibrosis.The results implied that Shahepatide had more research and application values on treating cirrhosis of the liver and chronic hepatitis.

从海洋生物鲨鱼肝脏纯化获得一种活性多肽—鲨肝肽 ,研究了其对CCl4 所致大鼠慢性肝损伤的治疗作用。结果表明 ,鲨肝肽对CCl4 引起的肝损伤大鼠血清中AST、ALT活性的升高、羟脯氨酸含量的升高均有抑制作用 ;能增加白蛋白含量 ,使白蛋白 /球蛋白比有一定的升高 ;肝组织病理切片观察显示 :鲨肝肽能显著减轻由CCl4 所致大鼠肝细胞脂肪变性及纤维组织增生。以上结果提示 :鲨肝肽有减轻肝脏纤维化的作用。在迁慢性肝炎、肝硬化等肝病治疗中具有良好的研究、应用前景。
